摘  要:【目的】探讨桃红四物汤联合诱导膜治疗大鼠骨缺损的疗效。【方法】将30只SD大鼠随机分为诱导膜+桃红四物汤组、诱导膜组、对照组等3组,每组10只。骨缺损模型是在大鼠股骨放置内固定钢板后截除4 mm的骨段形成骨缺损。对照组建立骨缺损模型,其余2组复制股骨骨缺损诱导膜模型。诱导膜组和诱导膜+桃红四物组模型建立后4周取出骨水泥并在骨缺损区植骨。从术后第2天开始,诱导膜+桃红四物汤组采用桃红四物汤剂灌胃治疗8周,诱导膜组及对照组组给予等体积生理盐水灌胃8周,术后12周取材,采用X射线摄片、苏木素-伊红(HE)染色法观察骨缺损区成骨情况,采用实时荧光定量逆转录聚合酶链反应(RT-PCR)法检测缺损植骨区血管内皮生长因子(VEGF)、骨形态发生蛋白2(BMP-2)、转化生长因子β1(TGF-β1)mRNA表达。【结果】诱导膜+桃红四物组Lane-Sandhu X射线评分,VEGF、BMP-2、TGF-β1 mRNA表达水平均高于诱导膜组及对照组(P <0.001)。【结论】桃红四物汤+诱导膜对骨缺损区的骨修复具有一定促进作用。Objective To observe the curative effect of Taohong Siwu Decoction combined with induced membrane for the treatment of bone defect in rats. Methods Thirty SD rats were randomly divided into three groups:induced membrane plus Taohong Siwu Decoction group,induced membrane group and control group,with 10 rats in each group. The bone defect model was formed by cutting off the bone segment of 4 mm after placing an internal fixation plate in the femur of rats. The bone defect model was established in the control group,and the induced membrane model of femoral bone defect was replicated in the other two groups. In the induced membrane group and the induced membrane plus Taohong Siwu Decoction group,bone cement was removed and bone graft was performed in the defect area 4 weeks after model establishment. From the second day after the operation, the induced membrane plus Taohong Siwu Decoction group was given intragastric administration of Taohong Siwu Decoction for8 weeks,the induced membrane group and control group were given intragastric administration of equal volume of normal saline for 8 weeks, and the samples were taken 12 weeks after the operation. X-ray radiography and hematoxylin-eosin(HE)staining were used to observe the osteogenesis in the bone defect area. The mRNA expression of vascular endothelial growth factor(VEGF), bone morphogenetic protein 2(BMP-2) and transforming growth factor β1(TGF-β1) in the bone defect area was detected by real-time fluorescence quantitative reverse transcription polymerase chain reaction(RT-PCR). Results The Lane-sandhu X-ray score,mRNA expression levels of VEGF,BMP-2 and TGF-β1 in the induced membrane plus Taohong Siwu Decoction group were higher than those in the induced membrane group and in the control group(P < 0.01). Conclusion Taohong Siwu Decoction combined with induced membrane is effective for promoting bone repair in bone defect area.
