摘  要:目的研究人参皂苷Rh2与肝癌细胞自噬的相关性,并探讨作用机制。方法免疫荧光法检测半胱氨酸蛋白酶-8/9(caspase-8/9)活性及自噬相关基因Beclin-1和LC3的表达。结果免疫荧光实验表明,20μg/mL人参皂苷Rh2作用肝癌HepG2细胞3 h,caspase-8活性和Beclin-1表达水平明显升高;caspase-9活性和LC3表达无显著变化。结论人参皂苷Rh2可以调节膜受体途径caspase-8和自噬相关基因Beclin-1的表达,发挥促进肿瘤细胞凋亡的生物学作用。Objective To study the correlation between ginsenoside Rh2 and autophagy of liver cancer cells,and to explore the mechanism of action.Methods Immunofluorescence method was used to detect the activity of caspase-8/9(caspase-8/9)and the expression of autophagy-related genes Beclin-1 and LC3.Results Immunofluorescence experiments showed that when 20μg/mL ginsenoside Rh2 acted on HepG2 cells for 3 h,caspase-8 activity and Beclin-1 expression levels were significantly increased;caspase-9 activity and LC3 expression did not change significantly.Conclusion Ginsenoside Rh2 can regulate the expression of membrane receptor pathway caspase-8 and autophagy-related gene Beclin-1,and play a biological role in promoting tumor cell apoptosis.
