摘  要:目的:研究推拿对CCI模型大鼠脊髓背角NMDA受体介导的下游CaMKⅡ和CREB表达的影响。方法:将40只SD大鼠平均分为5组,分别为空白组、模型组、推拿组、鞘内注射NMDA受体阻断剂组(MK-801)、鞘内注射阻断剂+推拿组,推拿组和鞘内注射阻断剂组均在造模后第4天进行干预和注射;HE染色观察脊髓背角神经细胞形态,Western Blot观察干预和鞘内注射后CaMKⅡ和CREB的表达情况。结果:HE染色显示,空白组与模型组相比,神经细胞形态有显著差异,但与阻断剂和阻断剂+推拿组相比差异不大;模型组与推拿组相比,并无显著性差异。Western Blot结果显示:推拿组、阻断剂组、阻断剂+推拿组CaMKⅡ和CREB表达量均降低。结论:推拿不能够改变腰膨大脊髓背角神经细胞的宏观形态,但推拿可能通过降低NMDA受体下游的CaMKⅡ和CREB表达量而起到镇痛作用。Objective:To investigate the effects of massage on NMD A receptor-mediated downstream CaMKⅡ and CREB expression in the spinal dorsal horn of CCI model rats.Methods:Forty SD rats were divided into 5 groups on average,which were blank group,model group,massage group,intraocular injection of NMD A receptor blocker group(MK-801),and intraocular injection of blocker+massage group.The massage group and intraocular injection blocker group were all treated with intervention and injection on the 4 th day after modeling.HE staining was used to observe the morphology of nerve cells in the dorsal horn of the spinal cord.Western Blot was used to observe the expression of CaMKII and CREB after the intervention and intrathecal injection.Results:HE staining showed that there was significant difference in the morphology of nerve cells between the blank group and the model group,but there was no significant difference between the blank group and the blocker and blocker+massage group;there was no significant difference between the model group and the massage group.Western Blot results showed that the expression levels of CAMK II and CREB were decreased in the massage group,the blocker group,and the blocker+massage group.Conclusion:Massage cannot change the macromorphology of spinal dorsal horn neurons in lumbar enlargement,but massage may play an analgesic role by reducing the expression of CaMKII and CREB downstream of NMDA receptor.
