摘 要:目的了解四川省急性弛缓性麻痹(acute flaccid paralysis,AFP)病例中分离的埃可病毒14型(echovirus 14,E14)的基因特征,探索其进化特征及在四川省的传播动态。方法从2006—2020年四川省AFP病例监测中分离鉴定到的11株E14毒株进行全VP1基因序列的测定及核苷酸同源性和遗传进化的分析。结果11株E14四川分离株分离于2006—2010年、2013—2014年,分布于四川省8个市。所有E14四川分离株核苷酸之间的同源性为81.2%~96.7%,氨基酸同源性为96.6%~99.6%。遗传进化树显示进化分支划分为4个簇cluster1~cluster4;四川分离株与中国山东省、云南省分离株同在cluster1,与其他簇的平均遗传距离为24.84%~26.18%,印度的大部分毒株在cluster3,与其他簇的平均遗传距离为22.81%~26.18%;澳大利亚的分离株和一小部分印度分离株聚在cluster4,与其他簇的平均遗传距离为22.81%~24.84%;原型株单独在cluster2,与其他簇的平均遗传距离为24.69%~25.30%。结论E14的进化较为活跃,地理分布上呈现一定聚集性,四川省内存在多条E14的传播链。Objective To understand the genetic characteristics of Echovirus 14(E14)isolated from acute flaccid paralysis(AFP)cases in Sichuan Province,and to explore its evolutionary characteristics and transmission dynamics in Sichuan province.Methods The whole VP1 gene sequences of 11 E14 strains which were isolated and identified from AFP case surveillance in Sichuan province from 2006 to 2020 were determined and analyzed for nucleotide homology and genetic evolution.Results 11 E14 Sichuan isolates were isolated in 2006-2010 and 2013-2014,and distributed in eight cities in Sichuan province.The homology among nucleotides of all E14 Sichuan isolates ranged from 81.2%to 96.7%,and the amino acid homology ranged from 96.6%to 99.6%.The genetic evolutionary tree showed that the evolutionary branching could be divided into four clusters;the Sichuan isolates were in cluster1 with isolates from Shandong province and Yunnan province in China,with an average genetic distance of 24.84%to 26.18%from other clusters,and most of the Indian strains were in cluster3,with an average genetic distance of 22.81%to 26.18%from other clusters;the Australian isolates and a small number of Indian isolates were clustered in cluster4,with an average genetic distance of 22.81%to 24.84%from other clusters;the prototype strain was alone in cluster2,with an average genetic distance of 24.69%to 25.30%from other clusters.Conclusion The evolution of E14 is more active,showing some aggregation in geographic distribution,and there are multiple transmission chains of E14 in Sichuan Province.
