摘  要:目的探究SMG6蛋白产物在主动脉夹层患者主动脉壁和正常主动脉壁中的表达情况及其表达变化的临床意义。方法选取2016年1月至2021年12月华中科技大学同济医学院附属同济医院心脏大血管外科连续收集的96份主动脉壁样本,通过Western Blot定量检测SMG6蛋白产物在主动脉壁中的表达水平,并分析其表达水平与主动脉夹层患者主动脉直径之间的相关性。结果 Western Blot结果显示,在主动脉壁中存在SMG6的两种蛋白异构体,分别是SMG6(57kDa)和SMG6(160kDa);在主动脉夹层患者的主动脉壁中,SMG6(57kDa)异构体的蛋白水平与正常对照组相比显著下降[(0.714±0.324)比(0.992±0.380),t=-3.1663,P=0.0029],而主动脉夹层动脉壁中SMG6(160kDa)异构体的蛋白水平与正常对照组相比则显著升高[(1.901±1.487)比(0.965±0.639),t=3.6173,P=0.00052],但是二者与主动脉直径的变化均无显著相关性。结论主动脉夹层发生过程中,SMG6的蛋白异构体表达发生显著变化,SMG6蛋白的不同异构体活性以及功能可能不同,通过干预SMG6的选择性剪切有望成为干预主动脉夹层的新靶点和新策略。Objective To explore the expression and clinical significance of SMG6 protein products in the ill aortic wall and normal aortic wall of patients with aortic dissection. Methods 96 samples of aortic wall from January 2016 to December 2021 were collected. The expression level of SMG6 protein products in aortic wall was quantitatively detected by Western Blot, and the correlation between the expression level and aortic diameter in patients with aortic dissection was analyzed. Results Western blot results showed that there were two protein isoforms of SMG6 in the aortic wall, SMG6(57 kDa) and SMG6(160 kDa). In the aortic wall of patients with aortic dissection, the protein level of SMG6(57 kDa) isoform(0.714 ± 0.324) was significantly lower than the normal control group(0.992 ± 0.380), while the protein level of SMG6(160 kDa) isoform in the aortic wall of patients with aortic dissection(1.901±1.487) was significantly higher than the normal control group(0.965±0.639).However, there was no significant correlation between them and the change of aortic diameter. Conclusions The expression of protein isomers of SMG6 changes significantly during the occurrence of aortic dissection, the activity and function of different isoforms of SMG6 protein may be different. Alternative splicing of SMG6 by intervention is expected to be a new target and strategy for the intervention of aortic dissection.
