摘 要:目的观察香青兰总黄酮对2型糖尿病(T2DM)大鼠模型的药效学研究。方法取动物10只做空白组,建模动物舌下静脉注射链脲佐菌素(STZ,60mg/kg)+高脂饲料喂养建立T2DM模型。选取建模成功动物依次分为模型组、盐酸二甲双胍组(167mg/kg)、香青兰总黄酮高剂量组(200mg/kg)、香青兰总黄酮低剂量组(100mg/kg)。各组动物(10只/组)分别给予相应药物灌胃,实验过程中检测各组动物动态血糖值、血清中胰岛素(INS)、胰岛素抗体(IAA)、肿瘤坏死因子(TNF-α)、白介素-1β(IL-1β)、白细胞介素-6(IL-6)、白介素-18(IL-18)、C-反应蛋白(CRP)、瘦素(LP)、脂联素(ADPN)、高迁移率族蛋白1(HMGB1)含量;分别摘取各组动物胰腺进行病理学检测。结果与模型组对比,香青兰总黄酮各剂量组动物血清空腹血糖值显著降低,IAA、TNF-α、IL-1β、IL-6、IL-18、CRP、HMGB1、LP指标含量也呈现降低趋势,而INS、ADPN含量呈现升高趋势(P<0.05或P<0.01)。结论香青兰总黄酮可降低T2DM动物空腹动态血糖值,修复受损胰腺,改善胰岛功能,作用机制可能与降低炎症反应有关。Objective To study the pharmacodynamics of total flavones from Cymbidium in type 2 diabetes mellitus(T2DM)rats.Methods 10 animals were taken as the blank group,and the T2DM model was established by sublingual intravenous injection of streptozotocin(STZ,60mg/kg)+high fat diet.The successful animals were divided into model group,metformin hydrochloride group(167mg/kg),high dose group of total flavonoids of vanillidium(200mg/kg),and low dose group of total flavonoids of vanillidium(100mg/kg).Animals in each group were given corresponding drugs by gavage.During the experiment,the dynamic blood glucose value of animals in each group was detected,and the serum was taken to detect insulin(INS),insulin antibody(IAA)and tumor necrosis factor(TNF-α)、Interleukin-1β(IL-1β)、Interleukin-6(IL-6),interleukin-18(IL-18),C-reactive protein(CRP),leptin(LP),adiponectin(ADPN),and high mobility group protein 1(HMGB1)contents.The pancreas of each group was extracted for pathological examination.Results Compared with the model group,the serum fasting blood glucose value in total flavonoid of Cymbidium was significantly reduced,and the levels of IAA,TNF-α,IL-1β,IL-6,IL-18,CRP,HMGB1,and LP were reduced,and the contents of INS and ADPN were increased(P<0.05 or P<0.01).Conclusion The total flavonoids of Cymbidium can reduce the fasting blood glucose level of T2DM animals,repair damaged pancreas,and improve islet function.The mechanism of action may be related to the reduction of inflammation.
