摘　　要：优选黄连小檗碱的最佳提取条件。采用乙醇回流提取法提取,考察过程要综合很多因素,提取时间与溶剂浓度都应划进考察范围,小檗碱含量为检测指标,用紫外分光光度法测定其含量。当盐酸小檗碱浓度保持在每毫升0.004~0.020 mg时,溶剂浓度同其吸光度存在线性关系,且呈现情况较为显著,线性方程为Y=49.993X+0.028 9,R^(2)=0.993,平均回收率为99.29%,用75%乙醇溶液提取3 h,黄连中小檗碱含量最高,达12.42%。优选黄连小檗碱的最佳提取条件为提取溶剂75%乙醇溶液,提取时间3 h。该提取条件可为中药黄连制剂生产提供一定的实验依据。In order to optimize the best extraction conditions of berberine in rhizoma coptidis, the method of ethanol reflux extraction was used, many factors were considered, which included extraction solvent and time.The content of berberine was taken as the testing index, its contents were determined by use of the UV-VIS.The experimental results showed that when the concentration of berberine hydrochloride was in the range of 0.004~0.020 mg·mL^(-1), the concentration and absorbance showed a good linear relationship, the linear equation was Y = 49.993 X +0.0289, R^(2)= 0.993, the average recovery was 99.29%, and when the extraction time was 3 h at 75% concentration of ethanol, the content of berberine in rhizoma coptidis was the highest, reaching to 12.42%. This condition can be the optimum extraction condition in the refluxing extraction of berberine in rhizoma coptidis.This study can provide a reference for the production of traditional Chinese medicines.
