葛根HPLC特征指纹图谱研究

摘　　要：采用高效液相色谱法建立了葛根药材的指纹图谱。使用Agilent Eclipse XDB-C_(18)色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.1%甲酸水(体积比)为流动相,建立了10批药材的指纹图谱共有模式。共标定了35个共有峰,并指认了其中5个色谱峰。各批次样品间相似度大于0.95。经主成分分析,各产地药材分为3类,与层次聚类分析结果一致。正交偏最小二乘法判别分析结果显示,共有15个共有峰VIP值>1,其中13号峰为葛根素,14号峰为葛根素-6″-O-木糖苷,15号峰为3′-甲氧基葛根素,16号峰为葛根素芹菜糖苷。本研究为葛根药材质量控制提供了参考。The HPLC fingerprint of Puerariae Raidx(PR)was established.Chromatographic separation was performed on an Agilent Eclipse XDB-C_(18) column(250 mm×4.6 mm,5μm).Acetonitrile and 0.1%formic acid-water(v/v)were used as the mobile phase for gradient elution.The HPLC fingerprint common pattern of 10 batches of PR was established.As a result,35 common peaks were determined and 5 of them were identified by comparing with reference substances.The similarity of 10 batches of samples was higher than 0.95.Based on PCA analysis,the samples were clustered into 3 categories,which was consistent with HCA results.The OPLS-DA results indicated that the VIP value of 15 common peaks were larger than 1.Among them,peak 13,14,15,16 were identified to puerarin,puerarin-6″-O-xyloside,3′-methoxypuerarin and mirificin,respectively.The experiment results provide reference for PR quality control research.
