摘 要:【目的】研究并初步建立石生茶藨的组培快繁体系,为石生茶藨种质资源的保存与繁育提供技术支持。【方法】以西天山野果林中重要伴生种石生茶藨的带芽茎段为外植体,分别以MS、B5和WPM培养基为基本培养基进行初代培养试验,以筛选出能促进石生茶藨腋芽萌发的最适培养基;分别以6-苄基腺嘌呤(6-BA)、吲哚丁酸(IBA)和赤霉素(GA_(3))为增殖培养的外源生长调节剂,采用二次回归正交旋转组合法进行增殖培养方案的设计,研究不同激素配比对芽分化、增殖及再生的影响情况;以1/2MS培养基为基本培养基,添加不同浓度的IBA,并进行不同时间的暗培养,以筛选出最适宜的生根培养基配方及暗处理时间。【结果】最能促进石生茶藨腋芽萌发的培养基是MS培养基,其萌发率可达91.11%;经二次多项式回归分析及以频率分析法模拟寻找最优值后发现,影响石生茶藨茎段增殖培养效果的植物生长调节剂的主次顺序是IBA>6-BA>GA3,其中IBA的使用对其增殖培养效果的影响显著,且IBA与6-BA的交互作用对其增殖培养效果的影响也显著,最适用的增殖培养基为MS+2.12 mg·L^(-1)6-BA+0.63 mg·L^(-1)IBA+0.66 mg·L^(-1)GA3,以此培养基进行增殖培养,可获得最大的增殖率;最适用的生根培养基为1/2MS+1.0 mg·L^(-1)IBA,以此培养基先暗培养3 d,其生根率可达86.67%。【结论】适用于石生茶藨组培快繁的初代培养基是MS培养基,IBA的使用对其增殖培养和生根培养均有显著效果,暗培养3 d可有效提高其生根率。【Objective】The tissue culture and rapid propagation system of Ribes Saxatile Pall.was preliminarily established to provide technical support for the preservation and breeding of R.Saxatile Pall.germplasm resources.【Method】The stems with buds of R.Saxatile Pall.,an important companion species in the wild fruit forest of Western Tianshan Mountains,were used as explants.The primary culture experiments were carried out with MS,B5 and WPM medium as the basic medium to screen the optimal medium for promoting axillary bud germination of R.Saxatile Pall..6-Benzyladenine(6-BA),indolebutyric acid(IBA)and gibberellin(GA_(3))were used as exogenous growth regulators for proliferation culture,respectively.The quadratic regression orthogonal rotation combination method was used to design the proliferation culture scheme,and the effects of different hormone ratios on bud differentiation,proliferation and regeneration were studied.Taking 1/2MS medium as basic medium,different concentrations of IBA were added and dark culture was carried out at different times to select the most suitable rooting medium formula and dark treatment time.【Result】The best medium to promote axillary bud germination of R.Saxatile Pall.was MS medium,which germination rate was 91.11%.Quadratic polynomial regression analysis and frequency analysis were used to simulate and find the optimal value.It was found that the primary and secondary order of plant growth regulators affecting the proliferation culture effect of R.Saxatile Pall.segment was IBA>6-BA>GA3.IBA had a significant effect on its proliferation culture effect,and the interaction between IBA and 6-BA also had a significant effect on its proliferation culture effect.The most suitable proliferation medium was MS+2.12 mg·L^(-1)6-BA+0.63 mg·L^(-1)IBA+0.66 mg·L^(-1)GA3.Proliferation culture with this medium could obtain the maximum proliferation rate;the most suitable rooting medium was 1/2MS+1.0 mg·L^(-1)IBA,and the rooting rate could reach 86.67%after dark culture for 3 days.【Conclu
