摘 要:为解决现阶段骨支架载药量难以控制、微球与多孔骨支架结合强度较低的问题,本研究利用挤出沉积式3D打印法和冷冻干燥法将丝素微球与羟基磷灰石支架相结合,制备出搭载异烟肼药物微球的复合支架。因为不同微球含量对支架性能有不同的影响,所以实验在定量控制支架内药物量的前提下,通过改变微球的含量研究支架的缓释、降解和力学性能。实验结果表明,丝素微球不仅能使羟基磷灰石复合支架的降解速度加快,而且可以延长释药时间和减少药物释放量,但微球含量的增加会一定程度上降低支架力学性能。因此,利用挤出沉积式3D打印法定量控制支架中载药微球含量,对微球与支架的结合、骨支架的药物控释具有良好的应用前景。It is known that the drug loading of bone scaffold is difficult to be controlled and the bonding strength between themicrospheres and theporous bone scaffolds is low.In order to solve the above problems,the silk fibroin microspheres arecombined with hydroxyapatite scaffolds using aextrusion deposition 3D printing method and afreeze-drying method,and composite scaffold containing isoniazid microspheres isprepared.The release,degradation and mechanical properties of scaffolds arestudied by changing the content of microspheres under the premise of quantitative control of drug amount in the stentsincethe different content of microspheres has different influences on the performance of scaffolds.The studyresults show that thesilk fibroin microspheres can not only accelerate the degradation rate of hydroxyapatite composite scaffolds,but also prolong the drug release time and reduce the drug release amount.However,with the increase of the content of microspheres,the mechanical properties of the scaffold arereduced to a certain extent.Therefore,the extrusion deposition 3D printing method can be used to quantitatively control the content of drug loaded microspheres in scaffolds,which has a good application prospect for the combination of microspheres and scaffolds and drug controlled release of bone scaffolds.
