摘 要:芦苇是山东省盐渍化地区常见的湿地盐生植物,对氮素转化以及氮去除有重要作用,其中微生物介导的反硝化过程被认为是主要的除氮机理。本研究以山东省不同盐渍化地区的芦苇根际土为材料,采用荧光定量PCR技术及高通量测序技术,对nirS、nirK及nosZ 3种基因型反硝化细菌的丰度以及nosZ基因型反硝化细菌的多样性、群落分布进行比较分析。结果表明,山东省盐渍化地区的芦苇根际中nosZ基因的拷贝数最高,昌邑和寿光地区的芦苇根际nirK基因拷贝数高于nirS,东营和滨州的芦苇根际nirS基因拷贝数高于nirK。固氮螺菌属(Azospirillum)是山东省盐渍化地区芦苇根际的优势反硝化细菌,尤其在昌邑(39.1%)和滨州地区(35.1%),Oligotropha是寿光芦苇根际丰度最高的反硝化菌(27.3%),假单胞菌属(Pseudomonas)是东营地区芦苇根际丰度最高的反硝化细菌(34.4%)。滨州地区的芦苇根际反硝化细菌多样性最高。昌邑和寿光两地的芦苇根际nosZ反硝化细菌群落结构相似,东营和滨州两地nosZ反硝化细菌群落组成差异较小。土壤的含盐量、Na+含量、含水量、总碳含量、铵盐以及pH值等环境因子均可影响芦苇根际nosZ基因型反硝化细菌的群落结构。Phragmites australis is one of the common wetland halophytes in salinized areas of Shandong Province.It plays imprortant roles in nitrogen transformation and removal.Denitrification process mediated by microbes is the main mechanism of nitrogen removal.In this study,with the rhizosphere soils of P.australis from different salinized areas of Shandong Province as research objects,the abundance of nirS,nirK and nosZ genes as well as the diversity and community distribution of nosZ-genotype denitrification bacteria were compared and analyzed using fluorescence quantitative PCR and high-throughput sequencing technology.The results showed that the copy number of nosZ gene was the highest,and that of nirK gene was higher than that of nirS gene in rhizosphere soil of P.australis from Changyi and Shouguang,while that of nirS gene was higher than that of nirK gene from Dongying and Binzhou.Azospirillum was the dominant denitrification bacterial species in rhizosphere soil of P.australis in salinized areas of Shandong Province,especially in Changyi(39.1%)and Binzhou(35.1%).Oligotropha had the highest abundance in rhizosphere soil of P.australis from Shouguang(27.3%),while Pseudomonas from Dongying(34.4%).The diversity of denitrifying bacteria in rhizosphere soil of P.australis from Binzhou was the highest.The community structure of nosZ genotype denitrification bacteria was similar between Changyi and Shouguang,and that between Dongying and Binzhou showed minor difference.The contents of salinity Na^(+)、soil moisture、total carbon、NH^(+)_(4)-N and pH value could affect the community structure of nosZ genotype denitrification bacteria in rhizosphere soil of P.australis.
