摘  要:目的研究阿托伐他汀对急性心肌梗死小鼠心肌组织赖安酰氧化酶表达的影响及机制。方法随机抽取8周龄健康清洁雄性C57BL/6小鼠30只,结扎冠状动脉前降支制作急性心肌梗死模型,随机分成两组,每组15只:(1)急性心肌梗死组(MI组:等量生理盐水灌胃);(2)治疗组(阿托伐他汀2 mg.kg^(-1).d^(-1)灌胃);另取15只健康清洁雄性C57BL/6小鼠设为对照组。第4周末处死小鼠,TUNEL染色检测细胞凋亡。提取左心室心肌组织RNA及蛋白质,实时荧光定量PCR法及Western Blot测定心肌组织LOX和MMP-2 mRNA和蛋白表达。结果TUNEL染色显示MI组小鼠心肌梗死周边细胞凋亡数增加,而治疗组小鼠心肌梗死周边细胞凋亡数减少。与对照组比较,MI组小鼠心肌组织LOX、MMP-2 mRNA和蛋白明显增加,差异有统计学意义(P<0.01);与MI组比较,治疗组小鼠心肌组织LOX、MMP-2 mRNA和蛋白明显下调,差异有统计学意义(P<0.01)。结论阿托伐他汀可下调急性心肌梗死小鼠心肌组织LOX表达,进而调控MMP-2表达。Objective To study the effects of atorvastatin on expression of lysyl oxidase(LOX)in myocardial tissue of mice with acute myocardial infarction(MI)and its mechanism.Methods Total of 30 C57BL/6 mice was randomly divided into 2 groups equally,the MI group was gastric lavage in normal saline,and treatment group was gastric lavage in atorvastatin 2 mg.kg^(-1).d^(-1) by ligtion of left anterior descending coronary artery for acute myocardial infarction model.Other fifteen normal mice were regarded as the control group.At the end of week 4,apoptosis was detected by TUNEL staining,expression level of LOX and MMP-2 mRNAs and proteins in myocardial tissue of MI mice was measured by real-time polymerase chain reaction(PCR)and western blot(WB)assay.Results TUNEL staining showed that the apoptosis of peripheral blood cells increased after myocardial infarction in MI group than that in the treatment group.The expression levels of LOX and MMP-2 mRNAs and proteins were significantly increased in MI group than that in control group(P<0.01),and were significantly decreased in treatment group than that in DCM group(P<0.01).Conclusion Atorvastatin can attenuate the increased-expression of LOX in myocardial tissue of MI rat,and then may regulate the expression of MMP-2.
