摘 要:开发了一种基于g-C_(3)N_(4)纳米片、铜离子(Cu^(2+))和邻苯二胺(OPD)的比率荧光探针,用于检测辅酶A(Coenzyme A,CoA)。分别采用g-C_(3)N_(4)纳米片和Cu^(2+)氧化OPD后的产物2,3-二氨基吩嗪(oxOPD)作为荧光指示剂,Cu^(2+)和oxOPD作为淬灭剂,检测体系中g-C_(3)N_(4)纳米片的荧光被淬灭,oxOPD荧光增强。CoA与Cu^(2+)形成络合物,减少了体系中作为氧化剂和淬灭剂的游离Cu^(2+)的含量,导致OPD的氧化过程受到抑制,oxOPD的含量减少,荧光强度降低,g-C_(3)N_(4)纳米片的荧光恢复。以g-C_(3)N_(4)纳米片的荧光发射强度与oxOPD的荧光发射强度比(F_(440)/F_(560))作为响应信号,用于比率荧光检测。在最佳实验条件下,采用本方法检测CoA,在1.0~100.0μmol/L浓度范围内具有良好的比率荧光响应,线性方程为F_(440)/F_(560)=0.00502C_(C oA)(μmol/L)+0.1079,R^(2)=0.984,检出限(LOD,3 S/m)为0.6μmol/L。此比率荧光检测体系具有良好的选择性,人血清中CoA的加标回收实验结果表明,此体系在复杂生物基质检测中具有良好的应用潜力。A detection system based on g-C_(3)N_(4)nanosheets,copper ions and o-phenylenediamine(OPD)was developed for detection of coenzyme A(CoA).By using chemically inert g-C_(3)N_(4)nanosheets and oxOPD as fluorescent indicators,CoA formed a complex with Cu^(2+),which reduced the amount of free Cu^(2+)as an oxidant and quencher in the system,resulting in the OPD oxidation process being affected.Inhibition reduced the amount of oxOPD that was used as a quencher and a fluorescent indicator,resulting in the recovery of the fluorescence of g-C_(3)N_(4)nanosheets,thereby changing the fluorescence intensity of the entire detection system.The ratio of the fluorescence emission intensity of g-C_(3)N_(4)nanosheets to that of oxOPD(F_(440)/F_(560))was used as the response signal for proportional fluorescence detection.Under the optimized experimental conditions,this method had a good proportional fluorescence response toward the concentration of CoA from 1.0μmol/L to 100.0μmol/L.The linear regression equation was F_(440)/F_(560)=0.00502 C_(CoA)(μmol/L)+0.1079(R^(2)=0.984).The detection limit(LOD=3 S/m)was 0.6μmol/L.The constructed detection system had good selectivity,and was used to detect CoA in human serum samples,which proved that the system had the potential for detection of CoA in complex biological matrices.
