摘 要:从甘蔗栽培种R570基因组数据库中挖掘到60个ShWRKY(ShWRKY1~ShWRKY600)基因,分布于10条染色体上,每条染色体上有3~13个ShWRKY基因.系统进化树分析表明60个ShWRKY蛋白中,属于Ⅰ、Ⅱ和Ⅲ类的分别有10、32和18个.所有ShWRKY家族成员皆为亲水性核定位蛋白,且除ShWRKY13为稳定蛋白外,其余家族成员都不稳定.ShWRKY基因家族的外显子数目为1~5个,保守基序数目为3~7个.启动子区域序列预测显示,ShWRKY基因包含多个与胁迫、生长发育和激素响应相关的顺式作用元件.表达谱分析显示,ShWRKY基因在甘蔗不同组织中差异表达,且参与对黑穗病菌胁迫的响应过程.RT-qPCR检测结果显示,接种黑穗病菌7 d后,ShWRKY37基因在2个甘蔗抗病品种和1个感病品种中上调表达,而在另外1个感病品种中下调表达;ShWRKY51基因则在抗病品种中下调表达,而在感病品种应答早期(1 d)上调表达.A total of 60 ShWRKY genes(ShWRKY1-ShWRKY60)were mined from the genomic database of Saccharum spp.hybrid cultivar R570.They were distributed on 10 chromosomes with 3-13 ShWRKY genes on each chromosome.Phylogenetic tree analysis showed that the number of ShWRKY proteins belonging to class Ⅰ,Ⅱ and Ⅲ was 10,32 and 18,respectively.All ShWRKYs were supposed to be hydrophilic and nuclear-localized,and unstable except that ShWRKY13 was a stable protein.The number of exons and conserved motifs within ShWRKY gene family were 1-5 and 3-7,respectively.Promoter region sequence prediction revealed that the ShWRKY genes contained multiple cis-acting elements which were related to stress,growth and development,and hormonal responses.Expression profile analysis indicated that the ShWRKY genes were differentially expressed in different tissues of sugarcane and participated in the regulation in response to Sporisorium scitamineum stress.RT-qPCR results revealed that after inoculation with S.scitamineum for 7 d,the expression of ShWRKY37 gene was up-regulated in 2 sugarcane resistant varieties and 1 susceptible variety,while down-regulated in another susceptible variety.Besides,the expression of ShWRKY51 was up-regulated in sugarcane resistant varieties and down-regulated in susceptible varieties in the early response stage(1 d).
