摘  要:为研究饲喂钩吻对仔猪肾脏的影响,将10头健康雄性去势二元杂种(长白猪×大白猪)断奶仔猪,随机分为对照组和给药组,每组5只.对照组仔猪饲喂不含抗菌药物的全价日粮,给药组仔猪饲喂不含抗菌药物的全价日粮+2%钩吻全草粉末,连续饲喂45 d后,分别采集两组猪的肾组织,对其进行高通量测序,并对差异表达基因进行基因本体论(GO)功能分类及京都基因和基因组百科全书(KEGG)数据库分析.结果显示,两组肾脏中共有301个差异表达基因.与对照组相比,给药组有180个基因显著下调,121个基因显著上调,其中与代谢、转运相关的基因,如溶质载体家族2成员13(SLC2A13)、溶质载体家族35成员5(SLC35A5)、含黄素单氧化酶2(FMO2)、醛氧化酶1(AOX1)等出现显著下调(P<0.05).荧光定量PCR对差异表达基因进行验证,其结果与转录组学结果基本一致.表明饲喂钩吻对仔猪肾脏的物质转运和药物代谢有一定影响.To study the effect of Gelsemium elegans supplementation on piglet kidney,10 healthy castrated two-way crossbred male piglets(Landrace×Yorkshire)were randomly divided into a control group and a medication group.In addition to complete formula feed,the medication group were continuously fed with 2%G.elegans powder for 45 d.Then the kidney tissues of all piglets were sampled and sequenced by high-throughput sequencing.The differentially expressed genes were classified by gene ontology(GO)function and analyzed using Kyoto Encyclopedia of Genes and Genomes(KEGG)database.A total of 301 differentially expressed genes were detected between the 2 groups.Compared with the control group,180 genes were significantly down-regulated and 121 genes were significantly up-regulated.Among them,the genes related to metabolism and transport,such as solute carrier family 2 member A13(SLC2A13),solute carrier family 35 member A5(SLC35A5),flavin containing monooxygenase 2(FMO2)and aldehyde oxidase(AOX1),were found to be down-regulated significantly(P<0.05),which were consistent with the results of real-time fluorescence quantitative PCR.In one word,G.elegans had some effects on the substance transport and drug metabolism in pig kidney.
