摘 要:目的探究三七通舒胶囊治疗脑缺血再灌注损伤大鼠的机制。方法于2019年1月—2020年12月间选取54只250~300 g雄性Wistar大鼠随机分成两组(手术后3天组及7天组),每组再分成3个亚组(假手术组、脑缺血再灌注损伤组和三七通舒胶囊治疗组),每组9只进行造模。对7天组大鼠每日评估神经功能。术后第3天和第7天终止实验,进行相对脑梗死体积测定,并通过免疫荧光和免疫印迹检测缺血神经元瞬时受体电位阳离子通道蛋白6(transient receptor potential cation channel 6,TRPC6)的表达水平。结果三七通舒胶囊治疗组大鼠神经功能在术后第4天开始较脑缺血再灌注损伤组大鼠显著改善(P<0.05,P<0.001),术后第3天及第7天相对脑梗死体积缩小(P<0.001),术后第7天TRPC6的表达水平显著增加(P<0.01)。结论三七通舒胶囊可以降低大鼠脑缺再灌注引起的神经功能损伤,其中可能的机制之一是其可以提高损伤神经元中的TRPC6表达水平。Objective The study is aimed to investigate whether Sanqi Tongshu Capsule(panaxatriol saponins,PTS)exerts its neuroprotective effects on ischemic stroke and whether its underlying mechanism through the TRPC6 pathway.Methods From January 2019 to December 2020,54 male Wistar rats with 250~300 g were randomly divided into two group on the grounds the time of reperfusion after ischemia(day 3 or day 7),then divided each group into three subgroups(n=9):(1)sham,(2)cerebral ischemia/reperfusion injury(I/R injury),(3)PTS treated(PTS+I/R injury)subjected to make experimental models.The neurological deficiency was assyed in the day 7 group.Relative infarct volume and TRPC6 expression detecting by immunofluorescence and Western blot analysis were assayed in the day 3 and day 7 group.Results Compared to the I/R injury group,the PTS+I/R injury group had a lower neurological severity scores from day 4(P<0.05,P<0.001),the relative infarct volume was significantly smaller in the day 3 and day 7(P<0.001),the protein expression of TRPC6 was significantly higher in the day 7(P<0.01).Conclusion Sanqi Tongshu Capsule can ameliorate the severity of nerve damage induced by cerebral I/R injury in rats and one of its mechanisms may be that it can increase the expression of TRPC6 in the impaired neurons.
