摘  要:目的探讨NDUFB2基因与心脏骤停后综合征(PCAS)脑损伤之间的关系。方法从GEO数据库下载表达谱数据GSE29540,通过生物信息方法进行目的基因筛选,将筛选出的基因在临床样本及细胞模型中进行验证。结果 GSE29540表达谱数据含有140例样本,其中神经功能预后良好(CPC 1~2分) 84例,神经功能预后不良(CPC 3~5分) 56例。通过生物信息学分析发现NDUFB2基因的预测价值最佳,因此将NDUFB2基因作为本研究的目的基因。临床水平及细胞水平验证均发现NDUFB2基因高表达与PCAS的神经功能恶化相关。结论 NDUFB2基因有望成为PCAS神经功能的生物标志物及潜在的干预靶点。Objective To study the relationship between NDUFB2 gene and the brain injury in post-cardiac arrest syndrome(PCAS).Methods Expression profile data GSE29540 was downloaded from GEO database.The target genes were screened by bioinformatics method.The selected genes were verified in the clinical samples and cell models.Results GSE29540 expression profile data included 140 samples,including 84 patients with good neurological prognosis(CPC 1-2 scores) and 56 patients with poor neurological prognosis(CPC 3-5 scores).According to bioinformatics analysis,we found that the predictive value of NDUFB2 gene was the best,so NDUFB2 gene was selected as the target gene in our study.High expression of NDUFB2 gene was found to be associated with the deterioration of neurological function in PCAS by both the clinical levels and the cellular levels.Conclusions The relationship between NDUFB2 gene and brain injury in post-cardiac arrest syndrome(PCAS) may be used as a biomarker of neurological function in PCAS and a potential intervention target.
