摘  要:【目的】试验旨在研究不同水平的葛根素对肉牛瘤胃体外发酵特性及养分降解率的影响。【方法】采集4头瘤胃瘘管牛瘤胃液,采用体外培养法,分别在基础日粮中添加0、0.01%、0.02%、0.04%、0.08%和0.16%的葛根素,检测体外培养24 h后瘤胃发酵指标和养分降解率的变化。【结果】葛根素各添加水平组的瘤胃液pH跟对照组相比无显著变化(P>0.05),菌体蛋白(MCP)浓度显著升高(P<0.05),0.02%、0.04%、0.08%和0.16%处理组的氨氮(NH3-N)含量和乙酸/丙酸均显著降低(P<0.05);各试验组的乙酸含量比对照组显著降低(P<0.05),丁酸和总酸含量均无显著变化(P>0.05);0.04%处理组的丙酸含量显著升高(P<0.05),其他处理组的丙酸含量无显著变化(P>0.05);此外,各试验组的干物质(DM)降解率均显著升高(P<0.05),0.04%、0.16%处理组的中性洗涤纤维(NDF)降解率以及0.04%、0.08%、0.16%处理组的粗蛋白(CP)降解率显著高于对照组(P<0.05)。【结论】添加葛根素后可促进瘤胃MCP的合成,提高瘤胃对饲料养分的降解率,调控瘤胃发酵模式。其中,以0.04%的剂量效果最好。[Objective]This experiment was conducted to investigate the effects of puerarin at different lev⁃els on rumen fermentation characteristics and nutrient degradability of beef cattle in vitro.[Method]Rumen flu⁃id was collected from 4 rumen fistulas cattle,with in vitro culture method,puerarin was added to the basal diet at the levels of 0,0.01%,0.02%,0.04%,0.08%and 0.16%,respectively.The changes in rumen fermentation in⁃dexes and nutrient degradability were detected 24 hours after in vitro cultur.[Results]There was no significant differences in rumen fluid pH between the puerarin group and the control group(P>0.05),the concentration of microbial protein(MCP)increased significantly(P<0.05),and the ammonia nitrogen(NH3-N)concentration and acetic acid/propionic acid in the 0.02%,0.04%,0.08%and 0.16%groups decreased significantly(P<0.05);the acetic acid concentration in all the experimental groups were significantly lower than that in the con⁃trol group(P<0.05),but the concentration of butyric acid and total acid did not change significantly(P>0.05);the propionic acid concentration in the 0.04%treatment group increased significantly(P<0.05),while the con⁃centration of propionic acid in other treatment groups had no significant differences(P>0.05);in addition,the degradability of dry matter(DM)in all the experimental groups increased significantly(P<0.05),the degradabil⁃ity of neutral detergent fiber(NDF)in the 0.04%and 0.08%puerarin groups and that of crude protein(CP)in the 0.04%,0.08%and 0.16%puerarin groups were significantly higher than that in the control group(P<0.05).[Conclusion]Puerarin can promote the synthesis of MCP,improve the rumen degradability of feed nutrients,and regulate rumen fermentation mode.The best dosage is 0.04%.
