摘 要:吸水链霉菌所产的子囊霉素(1)在工业上具有重要的应用价值,但存在遗传操作效率低等问题。本研究以1生产菌株吸水链霉菌SFK-6-33为研究对象,考察了影响吸水链霉菌与大肠埃希菌ET12567接合转移效率的因素,优化了吸水链霉菌与大肠埃希菌的接合转移遗传操作系统。结果表明,优化后的接合转移效率由最初的9.25×10^(–7)增加至4.33×10^(–5)。此外,在此基础上对吸水链霉菌生产1的相关调控基因(fkbN、fkbR1、shyT4和shyT10)进行研究,初步发现调控基因fkbN、shyT4能够促进1的合成,而调控基因fkbR1、shyT10对1的生物合成有抑制作用。Ascomycin(1) produced by Streptomyces hygroscopicus has important application value in industry,but it has problems such as low efficiency of genetic manipulation.Streptomyces hygroscopicus SFK-6-33 was used to investigate the factors affecting the efficiency of conjugal transfer between Streptomyces hygroscopicus and Escherichia coli ET12567,and the genetic operating system for conjugal transfer between Streptomyces hygroscopicus and Escherichia coli was optimized.The results showed that the optimized conjugation efficiency increased from the original 9.25×10^(–7) to 4.33×10^(–5).In addition,on this basis,the influences of relevant regulatory genes(fkbN,fkbR1,shyT4 and shyT10) of Streptomyces hygroscopicus on the production of 1 were investigated,and it was initially found that the regulatory genes fkbN and shyT4 could promote the synthesis of 1,while the regulatory genes fkbR1 and shyT10 were inhibitive.
