摘 要:目的探讨注射富血小板血浆(PRP)后大鼠光老化皮肤中Ⅰ和Ⅲ型胶原蛋白的表达量以及氧化应激反应。方法SD大鼠随机分为3组:正常对照组,模型组(紫外线照射大鼠背部皮肤14周+注射生理盐水4周),PRP治疗组(紫外线照射大鼠背部皮肤14周+注射PRP4周)。观察大鼠皮肤大体外观;Westernbolt法测定大鼠背部皮肤中Ⅰ和Ⅲ型胶原蛋白的含量;测定背部皮肤中MDA的含量和SOD的活性以评估氧化应激水平。结果与模型组比较,PRP治疗后:大鼠皮肤弹性明显好转,皱纹明显减少,无脱屑和明显色素沉着;皮肤中Ⅰ和Ⅲ型胶原蛋白的表达量较模型组明显增加;光老化皮肤中MDA的水平明显减少;SOD的活性增加。结论PRP治疗可以提高大鼠光老化皮肤中Ⅰ和Ⅲ型胶原蛋白的表达,并且抑制其氧化应激损伤。Objective To explore the expression of type Ⅰ and Ⅲ collagen and oxidative stress in the photoaging skin of rats after platelet-rich plasma(PRP) injection.Methods SD rats were randomly divided into 3 groups:normal control group(healthy rats),model group(ultraviolet irradiation on the back skin of rats for 14 weeks + injection with normal saline for 4 weeks),and PRP treatment group(ultraviolet irradiation on the back skin of rats for 14 weeks + injection with PRP for 4 weeks).The general appearance of skin was observed;the content of type Ⅰ and type Ⅲ collagen in the back skin of rats was measured by Western bolt method;the content of MDA and SOD activity in the back skin were measured to evaluate the level of oxidative stress.Results Compared with the model group,the skin elasticity of rats was significantly improved after PRP treatment,wrinkles were significantly reduced,there was no desquamation and obvious pigmentation;the expression of type Ⅰ and Ⅲ collagen in the skin was significantly increased compared with the model group;PRP treatment can reduce the level of MDA and increase the activity of SOD in photoaging skin.Conclusion PRP treatment can increase the expression of type Ⅱ and type Ⅲ collagen in photoaging skin of rats,and inhibit its oxidative stress damage.
