摘  要:目的用短串联重复序列(short tandem repeat,STR)基因分型法对人用疫苗检定用MRC-5和人横纹肌肉瘤细胞进行鉴定,确认无污染。方法选取人源细胞的20个STR位点进行等位基因PCR扩增及荧光检测,将得到的STR图谱与美国典型培养物保藏中心和德国微生物菌种保藏中心数据库比对分析。结果两株细胞均得到清晰的STR特征性图谱,未出现3单位基因,在两个数据库中均找到与其细胞分型完全匹配的细胞株。结论人用疫苗检定用MRC-5和人横纹肌肉瘤细胞均为正确细胞株,不存在污染和交叉污染。Objective To authenticate MRC-5 and human rhabdomyosarcoma(RD)cell lines used for human vaccine verification with short tandem repeat(STR)genotyping to ensure no contamination.Methods Twenty STR loci from human cells were selected for fluorescence detection after allele PCR amplification,and the obtained STR profiles were compared with American Type Culture Collection and German Collection of Microorganisms and Cell Culture databases.Results STR profiles were unambiguous and no triplicate peak appeared from either cell line.Cell lines that perfectly matched were found in both databases.Conclusion MRC-5 and RD cell lines used in human vaccine verification are correct cell lines without contamination or cross contamination.
