摘 要:目的:探究沙棘多糖对地塞米松诱导脂质代谢紊乱指标的作用,同时探究其对相关基因脂联素(Adiponectin)和低密度脂蛋白受体(LDLR)表达的影响。方法:选取健康成年雄大鼠35只,随机分对照组、模型组、低剂量组、中剂量组和高剂量组。通过腹腔注射地塞米松构建脂质代谢紊乱模型组;分别给低剂量组、中剂量组、高剂量组小鼠以沙棘多糖50、100、200 mg/kg的剂量灌胃,其余各组同体积生理盐水灌胃,灌胃持续4周。然后取大鼠血清检测总胆固醇(TC)、甘油三酯(TG)、血清游离脂肪酸(FFAs)、葡萄糖和Adiponectin的水平;利用Western Blot检测肝脏组织中LDLR的表达水平,并对结果进行统计学分析。结果:地塞米松诱导的模型组TC、TG、FFAs、葡萄糖水平升高;沙棘多糖处理下调了TC、TG、FFAs、葡萄糖水平;同时地塞米松能够降低血清中Adiponectin的水平,上调肝脏组织中LDLR的水平,沙棘多糖能够逆转这种作用。结论:沙棘多糖能够通过上调血清中Adiponectin的水平和下调肝脏组织中LDLR的表达,改善地塞米松诱导的脂质代谢紊乱。Objective:To explore the effect of seabuckthorn polysaccharide on dexamethasone-induced lipid metabolism disorder,and to explore its effect on the expression of related genes adiponectin and low-density lipoprotein receptor(LDLR).Methods:Thirty-five healthy adult male rats were randomly divided into control group,model group,low-dose group,middle-dose group,and high-dose group.The lipid metabolism disorder model group was established by intraperitoneal injection of dexamethasone;the low-dose group,middle-dose group and high-dose group were administered with seabuckthorn polysaccharide extract at a dose of 50,100,and 200 mg/kg,respectively,for 4 weeks,while the model group and control were intragastrically administered with the same volume of normal saline for 4 weeks.Rat serum was then extracted to detect the levels of blood lipids(TC,TG,and FFAs),glucose,and Adiponectin;Western Blot was adoped to detect the expression level of LDLR in liver tissue.The results were statistically analyzed.Results:After dexamethasone treatment(in model group),TC,TG,FFAs,and glucose levels increased,while Seabuckthorn polysaccharide treatment down-regulated these changes.Dexamethasone reduced serum Adiponectin levels and up-regulated LDLR levels in liver tissue,but Seabuckthorn polysaccharides could reverse these effect.Conclu-sion:Seabuckthorn polysaccharide can improve dexamethasone-induced lipid metabolism disorder by up-regulating the level of Adiponectin in serum and down-regulating the expression of LDLR in liver tissue.
