去除尼古丁和焦油的香烟烟雾提取物对大鼠阴茎平滑肌Cx43表达及勃起功能的影响

摘　　要：目的:探讨去除尼古丁和焦油的香烟烟雾提取物(fCSE)与抗氧化剂对阴茎平滑肌细胞缝隙连接蛋白Cx43表达及勃起功能的影响。方法:选30只SD雄性大鼠,随机分为3组:对照组(每日皮下注射PBS)、fCSE组(每日皮下注射fCSE)及fCSE+还原型谷胱甘肽(GSH)组(每日皮下注射fCSE+GSH)。8周后通过皮下注射脱水吗啡刺激后观察大鼠发生勃起的比例及次数;光镜下观察大鼠阴茎海绵体组织结构变化;利用激光共聚焦扫描显微镜观察阴茎海绵体平滑肌Cx43的表达。结果:fCSE组与对照组相比,大鼠阴茎勃起次数和勃起率显著下降(P<0.01),对照组与fCSE+GSH组无显著性差异(P>0.05);光镜下fCSE组较对照组平滑肌排列稍紊乱,血窦扩张,但未充血,纤维结缔组织较多,CSE+GSH组与对照组无明显差别;对照组和fCSE+GSH组Cx43密度分别达(65.00±5.57)%和(63.00±4.21)%,两者之间无统计学意义;fCSE组Cx43密度达(32.00±3.45)%,与对照组和fCSE+GSH组差异显著。结论:去除尼古丁和焦油的香烟烟雾提取物导致阴茎勃起功能下降,平滑肌Cx43密度下降,其机制可能是对连接蛋白造成了氧化应激损伤,影响到平滑肌细胞的增殖和迁移,进而影响到了勃起功能。Objective:To evaluate the effect of penile smooth muscle cells connexin 43(Cx43)density and erection of nicotine-and tar-free cigarette smoke extract(fCSE)in rats.Methods:Thirty SD male rats were randomly divided into control group(PBS group),fCSE group,and the fCSE+GSH group.After 8 weeks,rats were stimulated by subcutaneous injection of dehydrated morphine to observe the proportion and times of erectile.The histological changes of corpus erection rate of the experimental group decreased significantly(P<0.01),but there cavernosum were observed under light microscope and the expression of Cx43 in corpus cavernosum smooth muscle was observed by confocal laser scanning microscope.Results:The penile erection times and rates showed no significant difference between control group and antioxidant group(P>0.05).Under the light microscope,the smooth muscle arrangement of the experimental group was slightly disordered,the blood sinus was dilated but there was no congestion,and there were more fibrous connective tissue in fCSE group than in control group.There was no significant difference between antioxidant group and control group.Compared with that of control group and antioxidant group,the expression of Cx43 of the fCSE group were significantly lower,and their relative expression levels were(65.00±5.57)%,(63.00±4.21)%,and(32.00±3.45)%,respectively,P<0.05.Conclusion:Nicotine-and tar-free cigarette smoke extract results in the decrease in the density of Cx43 in rats′smooth muscle of penis and impairs their errect function,and the mechanism may be that connexin is damaged by oxidative stress,which affects the proliferation and migration of smooth muscle cells.
