摘 要:目的:探究锁阳乙酸乙酯提取物(ethyl acetate extract of cynomorium songaricum, ECS)对阿尔茨海默病(alzheimer disease, AD)小鼠肠道菌群紊乱的调节作用。方法:选取8周龄野生型C57BL/6小鼠为对照组,8周龄5xFAD转基因小鼠16只,分为模型组和ECS组,每组8只。适应性饲养1周后,ECS组小鼠灌胃47 mg·kg;ECS溶液,对照组和模型组分别灌胃等量去离子水,每天1次,连续84 d。采用Morris水迷宫(Morris water maze, MWM)实验检测小鼠的空间学习记忆能力;跳台实验检测小鼠的记忆能力;取小鼠肠组织内粪便样本进行16S rDNA测序,检测各组小鼠肠道菌群结构。结果:行为学结果显示,与对照组比较,模型组小鼠逃避潜伏期显著延长(P<0.05),穿越平台次数显著减少(P<0.05),跳台潜伏期明显缩短且错误次数显著增加(P<0.05);与模型组比较,ECS组小鼠逃避潜伏期显著缩短(P<0.05),穿越平台次数显著增加(P<0.05),跳台潜伏期显著延长且错误次数明显减少(P<0.05)。菌群检测结果显示:与对照组比较,模型组小鼠的菌群多样性降低,拟杆菌门、拟杆菌目、丹毒丝菌目、拟杆菌目S24-7及普雷沃氏菌科UCG-001丰度降低,厚壁菌门、梭菌目、乳杆菌属、毛螺菌科NK4A136丰度增加;与模型组比较,ESC组小鼠菌群多样性升高,拟杆菌门、拟杆菌目、丹毒丝菌目、拟杆菌目S24-7及普雷沃氏菌科UCG-001丰度显著上升,厚壁菌门、梭菌目丰度下降。结论:ECS可显著改善AD模型小鼠的空间记忆与学习能力,调节粪便菌群紊乱。Objective: To explore the regulatory effect of ethyl acetate extract of cynomorium songaricum(ECS) on intestinal flora disorder in Alzheimer′s disease(AD) mice.Methods: Eight week old wild-type C57 BL/6 mice were selected as the control group, and 16 8-week-old 5 xFAD transgenic mice were divided into model group and ECS group, with 8 mice in each group.After one week of adaptive feeding, mice in ECS group were gavaged with 47 mg/kg ECS solution, and mice in control group and model group were gavaged with the same amount of deionized water once a day for 84 days.Morris water maze(MWM) experiment was used to detect the spatial learning and memory ability of mice;the memory ability of mice was tested by platform jumping test;The fecal samples in mouse intestinal tissue were sequenced by 16 S rDNA,and the intestinal flora structure of mice in each group was detected.Results: The results showed that compared with the control group, the escape latency of the model group was significantly prolonged(P<0.05),the number of crossing the platform was significantly reduced(P<0.05),the platform jumping latency was significantly shortened and the number of errors was significantly increased(P<0.05);compared with the model group, the escape latency of ECS group was significantly shortened(P<0.05),the number of crossing the platform was significantly increased(P<0.05),the platform jumping latency was significantly prolonged and the number of errors was significantly reduced(P<0.05).The results of flora detection showed that compared with the control group, the flora diversity of model group mice decreased, the abundance of bacteroidea, bacteroidea, erysipelas, bacteroidea S24-7 and prevotellaceae UCG-001 decreased, and the abundance of Firmicutes, Clostridium, Lactobacillus and Spirillaceae NK4 A136 increased;compared with the model group, the flora diversity of mice in ESC group increased, the abundance of bacteroidea, bacteroidea, erysipelas, bacteroidea S24-7 and prevotellaceae UCG-001 increased significantly, and the abund
