摘 要:为了解广东地区某猪场猪流行性腹泻(PED)的流行情况,本试验通过病毒分离纯化、序列比对分析以确定疑似病料感染情况。结果显示:成功分离猪流行性腹泻病毒(PEDV),命名为GDsg株,该毒株属于变异株GIIa亚型分支,与经典株GIa亚型代表毒株CV777同源性最低,为96.6%;与经典株GIb亚型代表毒株SD-M同源性为97.0%;与变异株GIIa亚型代表毒株AJ1102同源性最高,为98.3%;与变异株GIIb亚型代表毒株CH-JX-1-2013同源性为98.1%。与经典株CV777相比,GDsg株S基因在中和抗原表位COE有6个氨基酸突变,在线性抗原表位SS6区间有1个氨基酸突变。本试验结果表明该猪场腹泻是由流行变异毒株GIIa亚型GDsg株引起的。This study isolated a porcine epidemic diarrhea virus(PEDV) strain(named GDsg strain) on Vero cells from small intestine samples of a suspected PEDV pig farm in Shaoguan city,Guangdong province,and showed that GDsg strain belonged to the GIIa subtype branch of the variant strain.Phylogenetic tree and homology alignment analyses revealed 96.6% nucleotide sequence homology with the classic strain GIa subtype representative strain CV777(lowest homology),97.0% nucleotide sequence homology with the classic strain GIb subtype representative strain SD-M,98.3% nucleotide sequence homology with the variant GIIa subtype strain AJ1102(highest homology),98.1% nucleotide sequence homology with the variant GIIb subtype strain CH-JX-1-2013.The amino acid sequence alignment indicated that the S gene of GDsg strain had 6 amino acid mutations in the neutralizing epitope COE and one amino acid mutation in the linear epitope SS6 in comparison to the classical strain CV777.These results suggested that GDsg strain,an endemic mutant strain GIIa subtype,was responsible for causing diarrhea in this Guangdong pig farm.
