摘 要:昆虫具有复杂而精细的嗅觉系统感知外界的气味信息。在嗅觉感受过程中,分布在触角或下颚须上的嗅觉感器通过表达多种蛋白完成气味的识别和转导。通过定位感器中的基因表达模式,可为研究嗅觉相关蛋白功能及互作机制提供必要的基础。本研究综述了RNA原位杂交技术在定位昆虫嗅觉相关基因的实验方法,分析了该技术在研究昆虫不同嗅觉基因定位和共定位中的应用;指出在进行昆虫RNA原位杂交研究时,应摸索出合适的实验条件和注重对照实验的设计以避免假阳性结果的出现。Insects have sophisticated olfactory system to perceive external odor information. In the process of olfactory sensation, the chemosensilla distributed on the antennae or maxillary palps can recognize and transduce the odor by expressing a variety of proteins. The locating of the gene expression patterns in the sensilla can provide a necessary basis for the study of olfactory related protein function and interaction mechanism. This article reviews the experimental methods of RNA in situ hybridization technology in locating insect olfactory-related genes, and analyzes the application of this technology in studying the locating of different olfactory genes in insects and the co-localization of two different genes. The article also points out that when conducting insect RNA in situ hybridization research, appropriate experimental conditions should be explored and the design of control experiments should be emphasized to avoid false positive results.
