GDF-15对糖尿病小鼠肝损伤保护作用的实验研究

摘　　要：目的探讨GDF-15对STZ所致C57BL/6J小鼠糖尿病肝脏损伤保护作用。方法将成模小鼠随机分为模型组、对照组及GDF-15注射组。监测血糖和体重。30 d后采集样本,检测各组小鼠血清中ALT、AST以及肝组织中TG、TC、SOD、MDA、HK及PK的表达。ELISA检测各组小鼠血清及肝组织中TNF-α、IL-1β、IL-6的表达。HE染色观察各组小鼠肝细胞形态,免疫组织化学染色和Western Blot检测各组小鼠肝组织中TNF-α、IL-1β、IL-6蛋白表达。结果与模型组比较,GDF-15组多饮、多食、多尿情况改善;体重升高,血糖降低;血清中ALT、AST水平降低;肝脏组织中TG、TC含量降低,SOD含量增高、MDA含量降低,HK、PK酶的活力增加。血清和肝组织中TNF-α、IL-1β、IL-6水平明显下降。肝细胞脂肪变性明显改善。免疫组织化学显示,TNF-α、IL-1β、IL-6在肝细胞中低表达。Western Blot显示,TNF-α、IL-1β、IL-6低表达。结论GDF-15对糖尿病肝损伤有一定的保护作用。GDF-15可能是通过抑制炎症反应减轻糖尿病肝损伤。Objective To investigate the protective effect of GDF-15 on liver injury in diabetic mice.Methods The model mice were randomly divided into model group,control group and GDF-15 injection group.Blood sugar and body weight were monitored.After 30 days,samples were collected to detect the expressions of ALT and AST in serum and TG,TC,SOD,MDA,HK and PK in liver tissue.The expressions of TNF-α,IL-1βand IL-6 in serum and liver tissues were detected by ELISA.HE staining was used to observe the morphology of liver cells in each group.Immunohistochemical staining and Western Blot were used to detect the expression of TNF-α,IL-1βand IL-6 proteins in liver tissues of each group.Results Compared with model group,the polydipsia,polyophagia and polyuria in the GDF-15 group were improved,with higher body weight and lower blood glucose;serum ALT and AST levels decreased;TG and TC levels in liver tissue decreased significantly;SOD content in liver tissue increased,while MDA content decreased significantly.The activities of HK and PK enzymes in liver tissue increased significantly.The levels of TNF-α,IL-1βand IL-6 in serum and liver tissue decreased significantly.Hepatocyte steatosis was significantly improved.Results of immunohistochemistry and Western Blot showed that the expressions of TNF-αIL-1βIL-6 in hepatocytes were low.Conclusion GDF-15 has some protective effect on diabetic liver injury and may alleviate diabetic liver injury by inhibiting inflammatory response.
