摘  要:目的探讨五味子素通过调控鼻咽癌细胞上皮间质转化(Epithelial-mesenchymal transition,EMT)过程影响其化疗敏感性的作用。方法取人鼻咽癌NPC-TW01细胞株进行细胞培养,分为A组(加不含五味子素及Notch激活剂Jagged1/FC培养液)、B组(加含1.2μg/ml Jagged1/FC培养液)、C组(加含10 mg/ml五味子素+1.2μg/ml Jagged1/FC培养液)、D组(加含10 mg/ml五味子素培养液)。采用四甲基偶氮唑盐比色法(MTT)法检测细胞增殖活性;Transwell小室法检测细胞侵袭能力;划痕实验法检测细胞迁移能力;CCK-8法检测药物半数抑制浓度(Half maximal inhibitory concentration,IC50),评估化疗敏感性;蛋白质印迹法检测Notch基因同源物1(Notch-1)、Notch胞内段(NICD)、Notch下游靶分子(Hes-1)蛋白相对表达量。结果与A组比较,B组、C组、D组各时段A值降低,且D组<C组<B组(P<0.05);与A组比较,B组、C组、D组侵袭细胞数减少,细胞迁移率、IC50值降低,且D组<C组<B组(P<0.05);与A组比较,B组、C组、D组Notch-1、NICD、Hes-1蛋白相对表达量降低,且D组<C组<B组(P<0.05)。结论五味子素可逆转鼻咽癌细胞EMT过程,增强化疗敏感性,作用机制可能与调节Notch信号通路有关。Objective To investigate the effect of Schisandrin to influence the chemotherapy sensitivity of nasopharyngeal carcinoma cells by regulating the epithelial-mesenchymal transition(EMT)process.Method Human nasopharyngeal carcinoma NPC-TW01 cell line was cultured and divided into group A(with Jagged1/FC medium without schisandrin and Notch activator)and group B(with 1.2μg/ml Jagged1/FC),group C(addition of 10 mg/ml schisandrin+1.2μg/ml Jagged1/FC culture solution),group D(addition of 10 mg/ml schisandrin culture solution).The tetramethylazolium salt colorimetric method(MTT)method was used to detect cell proliferation activity.The Transwell chamber method was used to detect cell invasion ability.The scratch test method was used to detect cell migration ability;the CCK-8 method was used to detect thehalf maxima inhibitory concentration(IC50)to evaluate chemotherapy sensitivity.Western blotting was used to detect the relative expression of Notch gene homolog 1(Notch-1),Notch intracellular segment(NICD),and Notch downstream target molecule(Hes-1).Result Compared with group A,the value of A in each period of group B,group C and group D decreased,and group D<group C<group B(P<0.05).Compared with group A,the number of invasive cells in group B,group C and group D decreased,and the cell migration rate and IC50 value decreased,and group D<group C<group B(P<0.05).Compared with group A,the expressions of Notch-1,NICD and Hes-1 in group B,C and D were decreased,and group D<group C<group B(P<0.05).Conclusion Schisandrin can reverse the EMT process of nasopharyngeal carcinoma cells and enhance the sensitivity of chemotherapy.The mechanism may be related to the regulation of Notch signaling pathway.
