乌萸汤改善小鼠卵巢储备功能及卵巢反应性的实验研究

摘　　要：目的通过观察卵巢储备功能下降(diminished ovarian reserve,DOR)小鼠促排卵情况及卵巢储备功能,探讨乌萸汤对DOR小鼠的治疗作用。方法以动情周期异常为DOR造模成功标准,将12只造模成功的24周龄C57BL/6J雌性小鼠随机分为高龄组和治疗组,6周龄C57BL/6J雌性小鼠为青年组。治疗组予乌萸汤灌胃8周,青年组和高龄组予等量蒸馏水灌胃。用药第56天起,小鼠腹腔序贯注射妊马雌酮及绒促性素,取卵进行体外受精实验观察其卵母细胞数、成熟卵数及受精卵数,血清抗苗勒管激素检测,卵巢HE染色观察卵巢内各级卵母细胞数量和质量、免疫荧光染色检测α-tubulin及Ki67表达。结果(1)治疗组小鼠血清抗苗勒管激素较高龄组有明显好转(P<0.05),且与青年组相比无统计学差异(P>0.05);(2)促排后,高龄组卵母细胞数、成熟率、受精率较青年组明显减少(P<0.05),治疗组与高龄组相比均有所升高(P<0.05);(3)高龄组卵巢略萎缩、髓质组织多,其内各级卵母细胞数量减少形态不佳,闭锁卵泡较多;治疗组髓质有所减少,生长卵泡细胞完整,形态有所好转;(4)治疗组小鼠卵巢α-tubulin及Ki67表达阳性率较高龄组有所升高(P<0.05)。结论乌萸汤能够改善DOR小鼠卵巢储备功能及卵巢低反应状态,从而提高生殖能力,具有一定的治疗作用。Objective To discuss the treatment effect of Wuyu decoction on mice with diminished ovarian reserve(DOR),by observing the ovulation induction and ovarian reserve function of DOR mice.Methods With abnormal estrous cycle as the criteria for successful DOR modeling,1224-week-old C57BL/6J female mice were randomly divided into the elderly group and the treatment group.The 6-week-old C57BL/6J female mice were divided into the young group.The treatment group was given Wuyu decoction by gavage for 8 weeks,and the young group and the old group were given the same amount of distilled water by gavage.From the 56th day,mice were injected intraperitoneally with estrone and choriotropin successively.The number of oocytes,mature eggs and fertilized eggs were observed by in vitro fertilization experiment,and the serum anti-mullerian hormone was detected.The number and quality of oocytes at all levels in ovary were observed by HE staining,and the expression levels ofα-tubulin and Ki67 were detected by immunofluorescence staining.Results(1)Compared with the elderly group,the serum anti-mullerian hormone in the treatment group was significantly improved(P<0.05),and there was no statistical difference between the treatment group and the young group(P>0.05).(2)After ovulation induction,the number of oocytes,the rate of mature eggs and fertilized eggs in the elderly group were significantly decreased than those in the young group(P<0.05),while those in the treatment group were increased compared with the elderly group(P<0.05).(3)In the elderly group,the ovary was slightly atrophy,medulla tissue was more,the number of oocytes at all levels was reduced and the morphology was not good,and there were more atresia follicles.In the treatment group,the medulla was reduced,the growth follicle cells were intact and the morphology was improved(P<0.05).(4)The positive expression rates ofα-tubulin and Ki67 in ovarian of mice in the treatment group were higher than those in the elderly group(P<0.05).Conclusion Wuyu decoction can improve the ova
