摘 要:目的探讨归术益坤方对多囊卵巢综合征(polycystic ovary syndrome,PCOS)大鼠卵巢功能的影响及其机制。方法SPF级雌性SD大鼠40只,随机分为空白组、模型组、中药高剂量组、中药低剂量组、阳性对照组,每组8只。除空白组外,每组均高脂饲料喂养联合丙酸睾酮注射液肌注,建立PCOS大鼠模型。造模2周后,中药高剂量组按1.8 g/100 g每日灌胃归术益坤方;中药低剂量组按0.9 g/100 g每日灌胃归术益坤方;阳性对照组按0.03 mg/kg每日灌胃炔雌醇环丙孕酮片混悬液;模型组及空白组给予每日2 mL生理盐水灌胃。给药3周后腹主动脉采血,血液离心后取血清行Elisa检测性激素;处死大鼠,剥离卵巢组织后称重并测量卵巢三径。左侧卵巢置于10%甲醛中固定,行石蜡切片;右侧卵巢冻存于液氮中备用,以Western Blot法检测SIRT1-FOXO通路相关蛋白。结果与空白组比较,模型组大鼠卵巢体积增大,卵巢相对质量增加,血清黄体生成素/卵泡刺激素比值升高,睾酮增高,SIRT1-FOXO通路相关蛋白(SIRT1、FOXO1、FOXO3、PPAR-α、PPAR-γ)表达升高。与模型组比较,中药高、低剂量组及阳性对照组均可降低血清黄体生成素/卵泡刺激素比值,降低血清睾酮水平,并下调SIRT1-FOXO通路相关蛋白水平,差异均有统计学意义(P<0.05)。其中阳性对照组的血清性激素水平改善优于中药高、低剂量组,而中药高剂量组对SIRT1-FOXO通路相关蛋白的调节效果优于阳性对照组。结论归术益坤方可改善PCOS大鼠卵巢形态学及性激素水平,其作用机制可能与对SIRT1-FOXO通路的调节作用有关。Objective To explore the effect of Guizhu Yikun formula on ovarian function and its mechanism on rats with Polycystic Ovary Syndrome(PCOS).Methods Forty SPF female SD rats were randomly divided into blank group,model group,high-dose group of Chinese medicinal,low-dose group of Chinese medicinal and positive group with 8 rats in each group.Except the blank group,each group was fed with high-fat diet combined with testosterone propionate injection to establish PCOS rats model.After two weeks,the high-dose group of Chinese medicinal was given 1.8 g/100 g Guizhu Yikun formula daily by intragastric administration,while the low-dose group was given 0.9 g/100 g.In the positive group,0.03 mg/kg ethinyl estradiol cyproterone tablet suspension was given by intragastric administration every day.The model group and blank group were given 2 mL normal saline every day.After three weeks of administration,blood was collected from abdominal aorta for blood centrifugation,and the serum was taken to detect sex hormones with Elisa method.Killed the rats and stripped the ovaries,then weighed and measured the three ovarian diameters.The left ovary was fixed in 10%formaldehyde for paraffin sectioned.The right ovary was frozen in liquid nitrogen and SIRT1-FOXO pathway related proteins were detected by Western Blot.Results Compared with the blank group,the volume and relative quality of ovaries in the model group increased.Serum luteinizing hormone(LH)/follicle stimulating hormone(FSH)ratio increased,testosterone increased,and SIRT1-FOXO pathway related protein expression increased.Compared with the model group,the high and low dose groups of Chinese medicinal and the positive group can reduce LH/FSH ratio,serum testosterone level and SIRT-FOXO pathway protein level,with statistical significance(P<0.05).The improvement of serum sex hormone level in the positive control group is better than that in the high-dose and low-dose groups of Chinese medicinal,while the regulation effect of the high-dose group of Chinese medicinal on SIRT1-FOXO p
