摘 要:目的基于网络药理学探究前列腺素类药物治疗糖尿病肾病(DKD)的作用机制。方法首都医科大学中日友好临床医学院肾病科通过Chemical Book平台、CTD数据库和SwissTargetPrediction服务器获取前列腺素类药物——前列地尔的基本信息和作用靶点;使用GeneCards和CTD数据库筛选DKD相关靶点。相关平台及数据库的数据更新截止于2021年3月。构建蛋白-蛋白相互作用(PPI)网络。对共同靶点进行基因本体(GO)及京都基因与基因组百科全书(KEGG)富集分析。结果共获得前列地尔作用靶点136个,DKD相关靶点5410个,交集靶点118个。PPI网络核心靶点25个。GO富集分析共得到1698个富集结果,主要涉及炎症反应、细胞增殖和凋亡等生物过程;KEGG富集分析共得到107条信号通路,主要涉及晚期糖基化终末产物(AGEs)-AGEs受体(RAGE)信号通路和低氧诱导因子-1(HIF-1)信号通路等。结论前列地尔等前列腺素类药物主要通过AGEs-RAGE和HIF-1信号通路,调控炎症反应、细胞增殖和凋亡等,延缓DKD进展。Objective To explore the mechanism of prostaglandins in the treatment of diabetic kidney disease(DKD)based on network pharmacology.Methods The basic information and targets of prostaglandin E1(PGE1,alprostadil)were obtained by Chemical Book,CTD and SwissTargetPrediction database,and DKD-related targets were obtained by searching GeneCards and CTD database,by the group from Department of Nephrology,China-Japan Friendship Clinical Medical College,Capital Medical University.the The data update of the relevant platforms and databases is as of March 2021.The protein-protein interaction(PPI)network was established.Gene Ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis of the common targets were conducted.Results A total of 136 targets of alprostadil and 5410 targets of DKD were obtained.The intersection of both were 118 targets.25 key targets are obtained by PPI network.GO enrichment analysis was carried out and 1698 results were obtained,including inflammatory response,cell proliferation and apoptosis,etc.Based on the enrichment analysis of KEGG,107 signaling pathways were obtained,including AGE-RAGE signaling pathway in diabetic complications,HIF-1 signaling pathways,etc.Conclusion Prostaglandins such as alprostadil can delay the progression of DKD through AGE-RAGE signaling pathway in diabetic complications and HIF-1 signaling pathway by regulating the inflammatory response,cell proliferation and apoptosis.
