摘 要:目的:探讨葡萄籽原花青素(GSP)对大鼠C6胶质瘤细胞活性氧、线粒体去极化和STAT3、P53、P21、Bcl-2 mRNA表达的影响。方法:采用水溶性四唑盐-1(WST-1)法测定GSP干预后C6胶质瘤细胞系的细胞活力,采用ROS指示剂荧光二氯荧光素二乙酸酯(H2DCFDA)测定干预后C6胶质瘤细胞内ROS水平,采用线粒体膜电位指示剂5,50,6,60-四氯-1,10,3,30-四甲基苯并咪唑碳氰化物碘化物(JC-1)测定干预后C6细胞的线粒体去极化,采用定量PCR法检测GSP干预后C6细胞的STAT3、P53、P21、Bcl-2 mRNA表达。结果:GSP各浓度处理C6胶质瘤细胞去极化细胞百分比、STAT3、P53、P21mRNAbiaoda水平均高于空白组(0 mg/L)(P <0.05),WST-1检测OD值、DCF-MFI和Bcl-2 mRNA水平均低于空白组(0 mg/L)(P <0.05);50 mg/LGSP处理C6胶质瘤细胞去极化细胞百分比、STAT3、P53、P21 mRNA水平均高于25 mg/L(P <0.05),WST-1检测OD值、DCF-MFI和Bcl-2 mRNA水平均低于25 mg/L;75 mg/LGSP处理C6胶质瘤细胞去极化细胞百分比、STAT3、P53、P21 mRNA水平均高于25、50 mg/L(P <0.05),OD值、DCF-MFI和Bcl-2 mRNA水平均低于25、50 mg/L。结论:GSP可以降低C6胶质瘤细胞内ROS水平,诱导线粒体去极化,抑制Bcl-2表达,诱导P21、STAT3和P53表达,抑制胶质瘤细胞增殖效呈剂量依赖趋势。Objective:To approach the effects of Grape Seed Proanthocyanidin( GSP) on rat C 6 glioma cells by observing the changes of ROS,mitochondrial depolarization and expressions of STAT3,Bcl-2,P53,P21 mRNA in C 6 glioma cells.Methods:The proliferation activity of C6 glioma cells was determined by WST-1,ROS was determined by H2 DCFDA,mitochondrial depolarization was determined by JC-1,and RT-PCR was used to examine the expressions of STAT3,Bcl-2,P53,P21 mRNA.Results:The percentage of mitochondrial de-polarization of C6 glioma cells,expressions of STAT3,P53,P21 mRNA in each concentration GSP group were higher than those of control group( P < 0.05),OD value,DCF-MFI and Bcl-2 mRNA expression of C6 glioma cells in each concentration GSP group were lower than those of control group( P < 0.05).Changes of all above indexes at concentration of 50 mg/L were more significant than those at concentration of 25 mg/L,above indexes at concentration of 75 mg/L were more significant than those at concentration of 25,50 mg/L.Conclusion:GSP can decrease ROS in C6 glioma cells,induce mitochondrial depolarization,inhibit Bcl-2 mRNA expression,increase STAT3,P53,P21 mRNA expressions,the inhibition on proliferation of C6 glioma cells was in a dose-dependent manner.
