海南黑山羊IGF2基因SNPs检测与生长性状的关联性分析

摘  要:以IGF2基因作为影响海南黑山羊生长性状的候选基因,采用直接测序和SnaPshot分型方法检测基因是否存在单核苷酸多态性,探讨不同基因型与海南黑山羊生长性状之间的相关性。结果只在IGF2基因第一外显子115 bp处检测到一个SNP多态性位点,突变位点G→A转换,属于同义突变,没有导致氨基酸改变。突变位点有效等位基因数(Ne)为1.166,观测杂合度(Ho)为0.135,期望杂合度(He)0.142,平均多态信息含量(PIC)为0.132,多态位点有三种基因型GG、GA、AA,GG基因型89个,GA基因型14个,AA基因型1个,基因型与与体长,体高,体重的关联性分析显示差异不显著。IGF2 gene was used as a candidate gene for growth traits of Hainan black goat.Single nucleo‐tide polymorphism(SNP)was detected by direct sequencing and snapshot genotyping to explore the correla‐tion between different genotypes and growth traits of Hainan black goat.The results showed that only one SNP polymorphism is detected at 115 bp of the first exon of IGF2 gene.The G→A transition of the muta‐tion site is a synonymous mutation and did not lead to amino acid changes.The effective number of alleles(Ne)is 1.166,the observed heterozygosity(Ho)is 0.135,the expected heterozygosity(He)is 0.142,and the average polymorphism information content(PIC)is 0.132.There are three genotypes of polymorphic loci:GG,GA,AA.There are 89 GG genotypes,14 GA genotypes,and 1 AA genotype.There is no significant dif‐ference between genotypes and body length,body height,and body weight.
