摘  要:本实验采用紫外分光光度法建立饲料中噁唑烷硫酮的定量检测方法。添加白芥子酶100 mg,饲料中的硫代葡萄糖苷在pH 7.0缓冲液中、35℃下酶解120 min,经二氯甲烷萃取后,带羟基的异硫氰酸酯与乙醇反应环化生成噁唑烷硫酮,测定试样溶液235、245 nm和255 nm的吸光度,紫外分光光度法定量。结果表明:所建立的方法定量限为150 mg/kg,平均回收率为86.1%~116.9%,相对标准偏差为1.27%~8.03%,方法重复性好,可信度高,适用于菜籽及其加工产品,以及含有菜籽及其加工产品的配合饲料、浓缩饲料和精料补充料样品中噁唑烷硫酮的测定。A quantitative determination method for oxazolidinethione in feeds by ultraviolet spectrophotometry was established.With 100 mg Sinapis alba L.enzyme hydrolysis,the glucosinolates in feeds were hydrolyzed at 35℃for 120 minutes.After extraction by dichloromethane,the hydroxy isothiocyanates were cyclized by ethanol,and converted into oxazolidinethione.The absorbance of test solution at 235,245 nm and 255 nm were determined.The oxazolidinethione content in feeds were quantitative determined by ultraviolet spectrophotometry.The results showed that the limit of quantification was 150 mg/kg.The average recovery were 86.1%~116.9%,and the relative standard deviations were 1.27%~8.03%.This method had good repeatability and high credibility,and could be used to determine the oxazolidinethione contents of rapeseed and its processed products,formula feed,concentrated and concentrated supplement.
