摘 要:目的探讨原花青素B2(procyanidin B2,PCB2)对PM_(2.5)急性暴露致大鼠心脏毒性的保护作用。方法24只雄性健康成年SD大鼠按照随机数字表法分为三组:生理盐水对照组(持续三蒸水灌胃21 d,在第8天、第10天、第12天气管滴注生理盐水)、PM_(2.5)组(持续三蒸水灌胃21 d,在第8天、第10天、第12天气管滴注PM_(2.5)悬浊液,10 mg/kg)、PCB2组(持续PCB2灌胃21 d,40 mg/kg,在第8天、第10天、第12天气管滴注PM_(2.5)悬浊液)。末次灌胃24 h后,处死大鼠,取心脏,制作组织切片,并检测心脏中总超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、丙二醛(malondialdehyde,MDA)、白细胞介素1β(interleukin-1β,IL-1β)、白细胞介素6(interleukin-6,IL-6)和白细胞介素18(interleukin-18,IL-18)的水平。结果与生理盐水对照组比较,PM_(2.5)组大鼠心脏组织匀浆中的IL-1β、IL-6、IL-18和MDA含量均明显升高,GSH-Px、SOD含量明显下降,差异均有统计学意义(P<0.05),与PM_(2.5)组相比,PCB2组大鼠心脏组织匀浆的IL-1β和IL-18含量均明显降低,SOD和GSH-Px含量升高,差异均有统计学意义(P<0.05)。结论急性气管滴注PM_(2.5)可明显引起大鼠心脏组织炎症和氧化应激指标改变,PCB2对PM_(2.5)引起的心脏损伤具有保护作用。Objective To explore the protective effect of procyanidin B2(PCB2)on cardiotoxicity induced by acute exposure to PM_(2.5) in rats.Methods A total of 24 healthy adult male SD rats were divided into three groups:the normal saline control group(the rats were given distilled water by gavage three time a day for 21 consecutive days and were exposed to 0.9%saline by intratracheal instillation on the 8th,10th,and 12th days),PM_(2.5) group(the rats were given distilled water by gavage three times a day for 21 consecutive days and were exposed to PM_(2.5) suspension by intratracheal instillation on the 8th,10th,and 12th days),and PCB2 group(the rats were pretreated with PCB2 by gavage for 21 consecutive days and were exposed to PM_(2.5) suspension by intratracheal instillation on the 8th,10th,and 12th days)according to random number table method.At 24 h after the last gavage,the rats were sacrificed and cardiac tissues were collected for prepartion of tissue section.The expressions of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),malondialdehyde(MDA),interleukin-1β(IL-1β),interleukin-6(IL-6)and interleukin-18(IL-18)were detected.Results Compared with the normal saline control group,the contents of IL-1β,IL-6,IL-18 and MDA in the heart tissue homogenate of PM_(2.5) group were significantly increased,while the contents of GSH-Px and SOD in PM_(2.5) group were decreased significantly,suggesting significant differences(P<0.05).Compared with the PM_(2.5) group,the contents of IL-1βand IL-18 in the heart tissue homogenate of PCB2 group were significantly reduced,and the contents of SOD and GSH-Px in PCB2 group were increased significantly,suggesting significant differences(P<0.05).Conclusion PM_(2.5) by acute intratracheal instillation can obviously cause inflammatory damage and oxidative stress changes in rats.PCB2 has a protective effect against heart damage caused by PM_(2.5).
