摘  要:目的:探究夏枯草提取物(PVE)对N-甲基-N’-硝基-N亚硝基胍(MNNG)诱导的大鼠胃癌组织Wnt/β-连接蛋白(β-catenin)信号通路的影响。方法:72只大鼠饮用水中添加MNNG,使其终浓度为100μg/ml,连续36周建立大鼠胃癌模型,将大鼠分为模型组、PVE(低、中、高)剂量组,每组18只,另18只健康大鼠设为对照组,饮用水为5%酒精水溶液。PVE(低、中、高)剂量组分别灌胃(0.5、1.0、2.0)g/kg PVE,对照组、模型组灌胃等体积生理盐水,每日1次,连续35d。观察大鼠饮食、精神状态、活动状态、粪便形状等一般情况;统计胃癌发生率及胃肿瘤体积大小;苏木精-伊红(HE)检测胃黏膜组织形态学;免疫组化检测胃癌组织中β-catenin、细胞周期蛋白D1(CyclinD1)蛋白表达情况。结果:模型组大鼠饮食量减少、两眼无神、活动量减少、粪便出现便溏现象,被毛松散蓬乱,色偏黄无光泽,胃黏膜不完整,出现部分断裂现象,胃壁弹性较差,基底膜的厚度不同,胃黏膜上皮细胞具有不同的形态大小和明显的异质性,增生腺体显著增加、侵入黏膜下层;PVE(低、中、高)剂量组出现不同程度的饮食量增加、精神状态渐佳、活动量增加现象,粪便逐渐成型,毛密集有光泽,胃黏膜不完整面积降低,断裂现象缓解,增生腺体侵入黏膜下层现象减弱。与对照组相比,模型组中胃癌发生率升高,胃肿瘤体积增大(P<0.05);与模型组相比,PVE(中、高)剂量组胃癌发生率,胃癌组织中β-catenin、CyclinD1蛋白阳性表达,PVE低剂量组胃癌组织中CyclinD1蛋白阳性表达降低(P<0.05),PVE(中、高)剂量组胃肿瘤体积减小(P<0.05);与PVE低剂量组相比,PVE高剂量组中胃癌发生率,胃癌组织中β-catenin、CyclinD1蛋白阳性表达,PVE中剂量组胃癌组织中β-catenin、CyclinD1蛋白阳性表达降低(P<0.05),PVE(中、高)剂量组胃肿瘤体积减小(P<0.05);与PVE中剂量组相比,PVE高剂量组胃癌组织中β-catenin、CyclinD1�Objective: To explore the effect of Prunella Vulgaris Extract(PVE) on Wnt/β-catenin signal pathway for rat gastric cancer tissue induced by N-methyl-N’-nitro-N-nitrosoguanidine(MNNG).Methods: MNNG was added to the drinking water of 72 rats to make the final concentration of 100μg/ml for 36 consecutive weeks to establish a rat gastric cancer model.The rats were divided into a model groups and PVE(low, medium, and high) dose groups, with 18 rats in each group, another 18 healthy rats were set as the controlled group and were given 5% alcohol aqueous solution for drinking.The PVE(low, medium, and high) dose groups were intragastrically administered(0.5,1.0,2.0) g/kg PVE,and the controlled group and model group were intragastrically administered with equal volume of normal saline, once a day for 35 consecutive days.The general conditions of the rat’s diet, mental state, activity state, and stool shape were observed;the incidence of gastric cancer and the size of gastric tumors were counted;the gastric mucosal tissue morphology was detected with hematoxylin-Eosin(HE);the protein expression of β-catenin and CyclinD1 in gastric cancer tissues was detected with immunohistochemistry.Results: The rats in the model group had reduced food intake, lost eyes, reduced activity, loose stools, loose, disheveled, yellow and dull coats, the gastric mucosa was incomplete, partially broken, and the gastric wall was less elastic, the thickness of the basement membrane was different, the epithelial cells of the gastric mucosa had different morphological sizes and obvious heterogeneity, and the hyperplastic glands increased significantly and invaded the submucosa;the PVE(low, medium, and high) dose groups showed varying degrees of increased diet, improved mental status, increased activity levels, the feces gradually formed, the coats were dense and shiny, the incomplete area of the gastric mucosa was reduced, the rupture phenomenon was relieved, and the intrusion of hyperplastic glands into the submucosa was weakened.Compared wit
