摘  要:目的观察电针额区对血管性痴呆大鼠认知功能及SIRT1/NF-κB信号通路的影响,初步探讨其作用机制。方法选取60只健康雄性Wistar大鼠,随机分为假手术组、模型组、西药组和电针组,动物模型的制作方法采取改良的2-VO法造模。电针组参照于致顺教授头针疗法,选取VD大鼠头部额区丛刺并连接脉冲电疗仪,西药组给予脑复康治疗,假手术组和模型组均给予生理盐水灌胃治疗。每组大鼠采用Morris水迷宫进行行为学能力的检测;Western blot法检测SIRT1,磷酸化NF-κB蛋白表达的变化。结果与模型组比较,电针组VD大鼠逃避潜伏期缩短(P<0.05),跨越原平台次数增高(P<0.05),VD大鼠海马区SIRT1蛋白表达升高(P<0.05),NF-κB p65蛋白表达明显降低(P<0.05)。结论电针额区可以有效改善VD大鼠的认知功能障碍,调整VD大鼠海马区SIRT1/NF-κB信号通路的传导,促进大鼠神经功能恢复。Objective To observe the ef fect of electroacupuncture in the frontal area on the cognitive function and SIRT1/NF-κB signaling pathway in rats with vascular dementia,and to preliminarily explore its mechanism.Methods A total of sixty healthy male Wistar rats were selected as the research subjects and randomly divided into a sham operation group,a model group,a western medicine group and an electroacupuncture group,and the animal model was established by the improved 2-VO method.In the electroacupuncture group,referring to Professor Yu Zhishun’s scalp acupuncture therapy,the frontal area of VD rats was manipulated with cluster scalp needling and connected to pulse electrotherapy device,while the western medicine group was given Naofukang,and the sham operation group and model group were treated with normal saline by gavage.Morris water maze was used to test the behavioral ability of rats in each group,and the changes in the SIRT1 and phosphorylated NF-κB protein expression were detected by Western blot.Results Compared with the model group,the escape latency of VD rats in the electroacupuncture group was shortened(P<0.05),and the number of crossing the original platform was increased(P<0.05),and the SIRT1 protein expression in the hippocampus of VD rats was increased(P<0.05),while the expression of NF-κB p65 protein was signifi cantly decreased(P<0.05).Conclusion Electroacupuncture in the frontal area can ef fectively improve the cognitive dysfunction of VD rats,adjust the conduction of SIRT1/NF-κB signaling pathway in the hippocampus of VD rats,and promote the recovery of neural function in rats.
