低农残高品质三七花总皂苷生产工艺研究

摘  要:目的结合现代生产工艺技术提取富集三七花总皂苷,同时脱除提取物中的农药和重金属残留,为开发该品种药品提供低残、健康安全的原料。方法前期通过小试正交实验,以三七花中人参皂苷Rb3、115项农药残留、四项重金属为检测指标,筛选出最佳工艺条件后进行三批中试放大。结果水作为提取溶剂,使用D101大孔吸附树脂进行富集,树脂:药材为2:1(m/m),用80%乙醇对总皂苷进行洗脱,洗脱流速为270 mL·min^(-1)~300 mL·min^(-1);洗脱液通过LK30树脂脱色,脱色流速为300 mL·min^(-1)~320 mL·min^(-1),脱色浓缩液中加入0.5%药材重量的活性炭吸附除杂质,最终浓缩浸膏使用浸膏重量3倍的丙酮结晶。结论通过此方法的应用,同时实现了三七花总皂苷的富集和总皂苷中农药、重金属残留的脱除,实验效果显著且该方法稳定,适用于现代工业生产。Objective To extract and enrich the total saponins in Radix Notoginseng flowers by combining modern production processes,and to remove pesticide and heavy metal residues in the extract,so as to provide low-residue,healthy and safe raw materials for the development of this variety of drugs.Methods In the early stage,a small-scale orthogonal experiment was conducted,using ginsenoside Rb3,115 pesticide residues and four heavy metals in Radix Notoginseng flowers as the detection indicators.After the optimal process conditions were screened out,three batches of pilot scale-up experiments were conducted using this process.Results Water was used as the extraction solvent and enriched with the D101 macroporous adsorption resin,with the ratio between the resin and medicinal materials as 2:1(m/m).After that,the extracted total saponins were eluted with80%ethanol at the flow rate of 270 mL·min^(-1)-300 mL·min^(-1).The eluent was decolorized by LK30 resin at a flow rate of 300mL·min^(-1)-320 mL·min^(-1).Activated carbon with the weight of 0.5%medicinal materials was added into the concentrated solution to adsorb and remove impurities.Finally,the concentrated extract was crystallized with acetone 3 times the weight of the extract.Conclusion The application of this production process fulfills the enrichment of total saponins in Radix Notoginseng flowers and the removal of pesticide and heavy metal residues from them simultaneously.The experimental results are significant and the method is stable,which is suitable for modern industrial production.
