摘  要:目的探讨葛根素对人鼻咽癌CNE-1细胞增殖及凋亡的影响,并探讨其与氧化应激的关系。方法以不同浓度葛根素(0、25、50、100μM)处理CNE-1细胞48 h,采用CCK-8法及流式细胞仪分别检测细胞增殖及凋亡,同时检测细胞中活性氧(reactive oxygen species,ROS)含量,丙二醛(malondialdehyde,MDA)含量及总超氧化物歧化酶(total superoxide dismutase,T-SOD)活性。结果不同浓度葛根素均可抑制CNE-1细胞增殖,诱导细胞凋亡,并增加细胞ROS及MDA含量,抑制T-SOD活性,且葛根素对CNE-1细胞的以上作用随其浓度的增加而增强。结论葛根素可抑制CNE-1细胞增殖,诱导细胞凋亡,作用机制可能与其促氧化应激有关。Objective To analyze the effect of puerarin on the proliferation and apoptosis of CNE-1 cells and to explore the association with oxidative stress.Methods CNE-1 cells were treated with different concentrations of puerarin(0,25,50,100μM)for 48 h.The proliferation and apoptosis of CNE-1 cells were respectively detected by CCK-8 assay and f low cytometry.The contents of reactive oxygen species(ROS)and malondialdehyde(MDA)and total superoxide dismutase(T-SOD)activity in CNE-1 cells were detected by the testing kit.Results Different concentrations of puerarin could inhibit proliferation and induce apoptosis of CNE-1 cells.Puerarin could increase the contents of ROS and MDA,and inhibit T-SOD activity of CNE-1 cells.These effects of puerarinon on CNE-1 cells were enhanced with the increase of its concentration.Conclusion Puerarin possesses the ability to inhibit the proliferation of CNE-1 cells and induce apoptosis,which may be related to its role in promoting oxidative stress.
