摘 要:目的观察葛根芩连汤对自发型2型糖尿病KKAy小鼠肠道菌群结构的影响。方法采用自发型2型糖尿病SPF级KKAy小鼠65只及C57BL小鼠13只,高脂饲料喂养,血糖>13.9mmol/L后,随机分为5组,正常组、阳性组(3.9mg/kg)、葛根芩连汤高、低剂量组(18.2、4.55g/kg)及模型组(生理盐水20mL/kg)。连续8周后留取粪便,采用16SrRNA检测菌群结构。结果因高脂饲料喂养,各给药组体质量均有不同程度上升,其中葛根芩连汤组有相对控制体质量上升的趋势。与模型组比较,各给药组均有不同程度降低血糖的作用(P<0.05、P<0.01)。同时,葛根芩连汤低剂量组及阳性组能够明显提高KKAy小鼠肠道菌群的群落丰度指数及多样性指数(P<0.01),包括OTU、ACE、chaol及shannon、simpson。在菌群结构方面,从纲、属、科水平发现葛根芩连汤能够提高Bacteroidia的丰度,降低Bacilli、Clostridia和Saccharimonadia丰度、占比。降低Lactobacillus丰度,提高Akkermansia和f_Muribaculaceae-Unclassified丰度。提示葛根芩连汤具有一定程度的类似于乳酸杆菌的调节作用,同时能够调节脂代谢达到缓解2型糖尿病及胰岛素抵抗的作用。结论葛根芩连汤能够改善糖尿病动物模型肠道菌群结构丰度及多样性,其作用机制可能是通过抑制慢性炎症水平,改善机体糖脂代谢,而达到改善2型糖尿病及胰岛素抵抗的作用。Objective To observe the effect of Gegen Qinlian Decoction on the intestinal flora structure of KKAy mice with spontaneous type 2 diabetes mellitus.Methods A total of 65 SPF KKAy mice and 13 C57BL mice with spontaneous type 2 diabetes mellitus were fed with a high-fat diet.After their blood glucose was higher than 13.9 mmol/L,they were randomly divided into 5 groups,e.g.normal group,positive group (3.9 mg/kg),Gegen Qinlian Decoction high-dose and low-dose groups (18.2,4.55 g/kg) and model group (normal saline 20 mL/kg).After 8 consecutive weeks,their feces were collected and their flora structures were detected by 16S rRNA.Results Due to the high-fat diet,the body mass of each administration group was increased to varying degrees,yet the Gegen Qinlian Decoction groups displayed a trend of relatively controlling the body mass increase.Compared with the model group,each administration group played the effect of reducing blood glucose in varying degrees (P<0.05,P<0.01).Meanwhile,the Gegen Qinlian Decoction low-dose group and the positive group significantly increased the community abundance index and diversity index of the intestinal flora of KKAy mice (P<0.01),including OTU,ACE,chaol,and shannon,simpson.In terms of the flora structure,it was found that Gegen Qinlian Decoction increased the abundance of Bacteroidia while reduced the abundance and proportion of Bacilli,Clostridia and Saccharimonadia,reduced the abundance of Lactobacillus yet increased the abundance of Akkermansia and f_Muribaculaceae_Unclassified on the level of class,genus,and family,suggesting that Gegen Qinlian Decoction had a certain degree of regulation effect similar to that of Lactobacillus,and at the same time regulated the lipid metabolism to alleviate type 2 diabetes and insulin resistance.Conclusion Gegen Qinlian Decoction can improve the abundance and diversity of the intestinal flora structure in diabetic animal models,whose mechanism may be performed through inhibiting chronic inflammation and improving the body’s glucose and lipid me
