摘 要:目的探究地石榴全草水提物的抗鸭乙型肝炎病毒(Duck Hepatitis B Virus,DHBV)效果及护肝作用。方法建立鸭乙型肝炎动物模型,将其分为6组:模型组、拉米夫定组、地石榴全草水提物高、中、低剂量组、空白组,每组10只,持续给药28 d。采用实时荧光定量PCR法测定给药前(T0)、给药后d7(T7)、d14(T14)、d21(T21)、d28(T28)和停止给药后d5(P5)的血清DHBV DNA水平,ELISA方法检测血清中DHBsAg和DHBeAg,试剂盒测定血清ALT、AST活性。最后取肝脏组织观察其病理改变。结果给药后地石榴全草水提物各剂量组的血清病毒载量和DHBsAg OD值均高于给药前(P<0.05或0.01),但尚不能确认地石榴水提物是否可以抑制DHBeAg。给药后地石榴全草水提物各剂量组的ALT活性和AST活性与给药前相比均有下降的趋势(P<0.05或0.01)。高剂量组的鸭肝损伤程度比模型组明显减轻(P=0.001)。结论地石榴全草水提物有一定的保肝降酶作用,但并未表现出抗DHBV功效。Objective To explore the anti-duck hepatitis B virus(DHBV)and liver protection effect of the aqueous extract of Melastoma dodecandrum Lour.Methods The animal model of duck hepatitis B was established and divided into 6 groups:model control group,positive drug group,high,medium and low dosage groups of water extract of Melastoma dodecandrum Lour and blank group,with 10 ducks in each group for 28 days.The serum DHBV DNA levels were measured by real-time fluorescence quantitative PCR before administration(T0),D7(T7),D14(T14),D21(T21),D28(T28)and D5(P5)after stopping administration.DHBsAg and DHBeAg in serum was detected by ELISA method while the activity of ALT and AST in serum were detected by kits.Finally,the liver was taken to observe the pathological morphology.Results After administration,the serum viral load and DHBsAg OD value of each dose group of the aqueous extract of Melastoma dodecandrum Lour were higher than those before administration(P<0.05 or 0.01),but it was not confirmed whether the aqueous extract of Melastoma dodecandrum Lour could inhibit DHBeAg.After administration,the ALT activity and AST activity of each dose group of the aqueous extract of Melastoma dodecandrum Lour showed a downward trend compared with those before administration(P<0.05 or 0.01).The degree of duck liver injury in the high-dose group was significantly less than that in the model group(P=0.001).Conclusion The aqueous extract of Melastoma dodecandrum Lour has a certain effect of protecting liver and reducing enzymes,but it does not show the effect of killing DHBV.
