摘 要:目的研究大黄素对高氧肺损伤的保护作用及机制。方法采用高氧暴露法(85%)建立新生SD大鼠支气管肺发育不良(bronchopulmonary dysplasia,BPD)模型,测定大鼠体质量和肺组织含水量,对肺组织标本进行组织学分析,酶联免疫吸附法(ELISA)检测血清中IL-1β、IL-8、TNF-α改变,Western blot检测核因子-κB(NF-κB)p65表达变化。结果HE染色显示高氧抑制肺泡发育,肺泡数量减少,肺泡间隔增加,肺组织毛细血管形态不规整;大黄素治疗组大鼠体重较模型组增加(P<0.05),肺组织含水量较模型组降低(P<0.05);外周血IL-1β、IL-8、TNF-α含量增加,与模型组比较,大黄素组大鼠体质量增加(P<0.05),肺组织含水量明显降低(P<0.05),外周血IL-1β、IL-8、TNF-α含量降低;模型组大鼠肺组织NF-κB p65表达升高,大黄素组NF-κB p65表达下调(P<0.05)。结论大黄素通过抑制炎症反应对新生大鼠肺组织产生保护作用。Objective To study the protective effect of emodin on hyperoxia induced lung injury and its mechanism.Methods The model of bronchopulmonary dysplasia(BPD)was established by high oxygen exposure(85%),the mass and water content of lung tissue were measured,the lung tissue samples were analyzed histologically,the changes of IL-1β,IL-8 and TNF-αin serum were detected by enzyme-linked immunosorbent assay(ELISA),and the expression of nuclear factor-κB(NF-κB)p65 was detected by Western blot.Results HE staining showed that under hyperoxia the development of alveoli was inhibited,the number of alveoli was decreased,the alveolar septum was increased,and the shape of pulmonary capillaries was irregular;Compared with the model group,the weight of rats in emodin treatment group increased(P<0.05),and the water content of lung tissue decreased(P<0.05),and the contents of IL-1β,IL-8,TNF-αin peripheral blood increased;Compared with the model group,body weight of rats in emodin group increased(P<0.05),lung water content decreased significantly(P<0.05),and peripheral blood IL-1β,IL-8 and TNF-αcontents decreased;The expression of NF-κB p65 in lung tissue increased in model group,but decreased in emodin group(P<0.05).Conclusion Emodin could inhibit inflammation and protect lung tissue of newborn rats.
