哈尔滨市中老年人膝关节骨性关节炎的靶标代谢组学研究

摘  要:目的探讨哈尔滨市中老年人膝关节骨性关节炎患者体内脂肪酸和氨基酸的代谢变化。方法选取于我院拟行膝关节置换手术的关节炎患者76例和非关节炎患者71例作为研究对象;采集患者血清样本,应用靶标代谢组学分析方法检测膝关节骨性关节炎患者的血清脂肪酸和氨基酸含量的变化;运用多变量统计分析方法筛选出膝关节骨性关节炎患者的敏感、可靠的生物标志物。结果与对照组比较,膝关节骨性关节炎患者血清中分别检测到10种脂肪酸和21种氨基酸出现异常变化(P<0.05)。综合变量重要性投影和载荷图的结果显示二十二碳六烯酸、亮氨酸、4-羟基-L-脯氨酸、苯丙氨酸、丙氨酸可作为敏感的生物标志物。结论哈尔滨市中老年人膝关节骨性关节炎血清脂肪酸和氨基酸含量发生异常,这些结果可能会为骨性关节炎的诊断和治疗提供新的思路。Objective To investigate the metabolic changes of fatty acids and amino acids in middle-aged and old patients with knee osteoarthritis in Harbin.Methods Seventy-six patients with arthritis and 71 patients with non arthritis were selected as the research objects.The changes of serum fatty acids and amino acids in patients with knee osteoarthritis were detected by target analysis,and the sensitive and reliable biomarkers were screened out by multivariate statistical analysis.Results Compared with the control group,the abnormal changes of 10 fatty acids and 21 amino acids were found in the serum of patients with knee osteoarthritis(P<0.05).The biomarkers were confirmed by the results of VIP and load map.It was found that docosahexaenoic acid,leucine,4-hydroxy-l-proline,phenylalanine and alanine could be used as sensitive biomarkers for the diagnosis of knee osteoarthritis.Conclusion The content of fatty acids and amino acids in serum of middle-aged and old people with knee osteoarthritis in Harbin is abnormal,which may provide new ideas for the diagnosis and treatment of osteoarthritis.
