自拟消瘤方对实验性大鼠子宫肌瘤的作用机制研究

摘　　要：目的探讨自拟消瘤方防治大鼠子宫肌瘤的分子作用机制。方法以己烯雌酚和黄体酮负荷法诱导建立实验性SD大鼠子宫肌瘤模型,观察自拟消瘤方对动物子宫系数,子宫湿重及子宫平滑肌病理改变的影响;酶联免疫法分析自拟消瘤方对血清雌二醇和孕酮水平的影响;免疫组化法观察自拟消瘤方对大鼠子宫雌激素受体(ER)、孕激素受体(PR)的影响。结果与模型组对比,自拟消瘤方不同干预组子宫系数和子宫湿重变化呈剂量依赖性降低(P<0.01)。组织病理学观察发现自拟消瘤方干预组的大鼠子宫平滑肌肌层增厚减缓,肌层内细胞排列有序,增生范围皱缩,胞核椭圆状向梭状转变。自拟消瘤方各干预组的雌二醇和孕酮水平均有明显下降(P<0.05),免疫组化结果显示自拟消瘤方干预后能够明显下调ER、PR的表达(P<0.01)。结论推测自拟消瘤方可能通过下调雌激素和孕激素的表达从而发挥防治大鼠子宫肌瘤的作用。Objective To study the effect of self-designed Xiaoliu prescriptionon uterine fibroids of experimental ratsand to explore its mechanism.Methods The experimental model of uterine fibroids in experimental SD rats was induced by diethylstilbestrol and progesterone loading method.The effects of self-designed Xiaoliu prescriptionon uterus and ovary,uterine wet weight and pathological changes of uterine smooth muscle were observed.The effect of self-designed Xiaoliu Recipe on serum estradiol and progesterone levels was analyzed by immunohistochemical method.Immunohistochemical method was used to observe the effect of Xiaoliu recipe on uterine estrogen receptor(ER)and progesterone receptor(PR)in rats.Results Compared with the model group,the uterine coefficient and wet weight of the uterus in the different intervention groups of the self-designed Xiaoliu Recipe decreased in a dose-dependent manner(P<0.01).Histopathological observation showed that the increase of thickness atthe uterine smooth muscle layer of rats in the intervention group of the self-designedXiaoliu Recipe was slowed down,the cells in the muscle layer were arranged in an orderly manner,the proliferation area was shrunk,and the nucleus was elliptical to spindle-shaped.The levels of estradiol and progesterone in each intervention group of the self-designed Xiaoliu Recipe decreased significantly(P<0.05).The immunohistochemical results showed that the self-designed Xiaoliu Recipe can significantly down-regulate the expression of ER and PR(P<0.01).Conclusions Self-designed Xiaoliu prescription has a certain therapeutic effect on experimental rat uterine fibroids,and its effect may be achieved by reducing the expression of ER and PR.
