摘 要:以枯否氏细胞(KCs)为细胞模型,研究miR-14对麦冬多糖脂质体(OPL)调节免疫活性的影响。首先转染miR-14 mimic或miR-14 inhibitor,再用OPL处理,然后采用Griess、流式细胞术、荧光染色以及细胞划痕等方法检测NO和iNOS的分泌、吞噬FITC-dextran和FITC-OVA的活性、表面分子CD14和MHCⅡ的表达、细胞凋亡、ROS分泌。结果显示,转染miR-14 inhibitor后,OPL能够显著促进NO的分泌和MHCⅡ的表达;转染miR-14 mimic和miR-14 inhibitor后,OPL能够显著提高KCs的吞噬活性,促进iNOS和CD14的表达,并能够降低细胞凋亡水平和ROS的分泌。结果表明,OPL能够通过调控miR-14的表达来提高KCs的免疫活性。To study the effects of miR-14 on regulation of immune activity by Ophiopogon polysaccharide liposome(OPL)on Kupffer cells(KCs),KCs used as the cell model were transfected with miR-14 mimic or miR-14 inhibitor and then treated with OPL.Griess,flow cytometry,fluorescence staining and cell scratch were used to detect NO and iNOS productions,phagocytic activity of FITC-dextran and FITC-OVA,surface molecules CD14 and MHCⅡexpressions,cell apoptosis and ROS.The results from KCs transfected with miR-14 inhibitor showed that OPL could increase the generation of NO and upregulate the expressions of MHCⅡ.In addition,the results from KCs transfected with miR-14 mimic and miR-14 inhibitor showed that OPL could improve the phagocytic activity of KCs,increase the expressions of iNOS and CD14,reduce the apoptosis level and ROS secretion.The study demonstrated that OPL could improve the immune activity of KCs by regulating the expression of miR-14.
