摘 要:目的基于网络药理学方法分析葶苈大枣泄肺汤治疗结核性胸膜炎的作用机制。方法借助中药系统药理学分析平台(TCMSP)和蛋白数据库(UniProt)获得葶苈子、大枣的活性成分及其作用靶点。利用Genecards数据库和OMIM数据库获取结核性胸膜炎相关疾病靶点,并与活性成分靶点进行映射,取交集,得到葶苈大枣泄肺汤治疗结核性胸膜炎的共同(直接)靶点。使用String数据库和Cytoscape 3.7.2软件构建靶蛋白互作(PPI)网络及活性成分-靶点网络。通过Metascape数据库对共同靶点进行GO功能和KEGG通路富集分析,借助微生信网站绘制富集气泡图,利用Cytoscape 3.7.2软件构建共同靶点-信号通路网络。结果共有18个活性成分通过24个共同靶点作用于结核性胸膜炎,核心靶点主要包括MMP9、VEGFA、IL6、TNF、IL1B、CCL2等。共同靶点的GO功能富集分析涉及包括细胞因子信号转导、趋化因子调控、脂多糖应答在内的主要生物进程。KEGG通路富集分析得到AGE-RAGE、NF-κB、HIF-1、癌症中的转录失调、癌症中的蛋白聚糖通路等11条显著相关的信号通路。结论葶苈大枣泄肺汤可能主要通过抑制炎症、增强免疫、抗氧化等途径对结核性胸膜炎起到治疗作用。Objective To analyze the therapeutic mechanism of Tingli Dazao Xiefei decoction on tuberculous pleurisy using method of network pharmacology.Methods The active components,target proteins and target genes of Descurainiae semen lepidii semen and Jjujubae fructus were searched by TCMSP and Uniprot database.These genes were mapped with the tuberculous pleurisy related genes obtained by Genecards and OMIM database,and were selected as the common target genes(direct targets)for the treatment of tuberculous pleurisy with Tingli Dazao Xiefei decoction.String database and Cytoscape 3.7.2 software were used to construct the protein-protein interaction(PPI)network and the active ingredient-core target network.Then,GO and KEGG enrichment analysis of core target genes were carried out through Metascape database,and enrichment bubble map was drawn with the help of Microbioinformatics website.Finally,we used Cytoscape 3.7.2 software to construct the network of core target-signal pathway.Results 18 active components could act on tuberculous pleurisy through 24 core targets,and core target genes were mainly involved in MMP9,VEGFA,IL6,TNF,IL1 B,CCL2,etc.The GO(BP)enrichment analysis of core targets identified major biological processes including cytokine signal transduction,chemokine regulation and lipopolysaccharide response.Enrichment analysis of KEGG pathway revealed 11 signal pathways with significant correlation,including AGE-RAGE,NF-κB,HIF-1,transcription disorders in cancer,and proteoglycan pathway in cancer.Conclusion Tingli Dazao Xiefei decoction may treat tuberculous pleurisy by inhibiting inflammation,enhancing immunity,anti-oxidation and other pathways.
