摘  要:随着转基因作物商品化和基因组编辑技术的迅速发展,植物基因工程安全性越发引人关注。最常用的转基因植物筛选标记一般是抗生素抗性基因或者除草剂抗性基因,存在安全性和成本问题。通过前期研究发现,乙酰乳酸合成酶(Acetolactate synthase,ALS)基因突变型OsALS S627N具有咪唑啉酮类除草剂抗性,推测该基因能用于转基因植物筛选。通过构建该基因植物表达载体转化拟南芥,潮霉素、除草剂筛选和GUS染色检测,发现OsALS S627N确实能有效筛选出阳性转基因植株。OsALS S627N来源于植物且动物不含有该基因、无抗生素抗性基因转移风险且咪唑啉酮类除草剂成本低,因此能够作为一个新的安全有效的转基因植物筛选标记。With the commercialization of transgenic crops and the rapid development of genome editing technology,the safety of plant genetic engineering has attracted more and more attention.The most commonly used transgenic plant screening markers are antibiotic resistance genes or herbicide resistance genes,which have safety and cost problems.Our previous research found that the mutant OsALS S627N of acetyllactate synthase(ALS)gene has imidazolinone herbicide resistance.It is speculated that this gene can be used for the screening of transgenic plants.By constructing the plant expression vector of the gene and transforming it into Arabidopsis,hygromycin,herbicide screening and GUS staining,it was found that OsALS S627N could effectively screen positive transgenic plants.Os-ALS S627N comes from plants and animals do not contain this gene,there is no risk of antibiotic resistance gene transfer,and the cost of imidazolinone herbicides is low.Therefore,OsALS S627N can be used as a new safe and effective transgenic plant screening marker.
