摘 要:目的:探究维吾尔药祖卡木颗粒对大鼠急性肺损伤模型抗炎作用及其可能的作用机制。方法:适应性饲养3 d后将48只大鼠按随机数字表法随机分成对照组、模型组、阳性药物组和低、中、高剂量祖卡木组6组,祖卡木低、中、高剂量组分别给予1.35 g/kg、2.7 g/kg和5.4 g/kg灌胃干预5 d,阳性药物组于实验进行的第5天给予腹腔注射地塞米松(5 mg/kg),对照组与模型组给予等体积生理盐水处理,于给药干预结束后的1h采用脂多糖进行大鼠急性肺损伤模型构建。苏木精-伊红染色法观察大鼠肺组织的病理损伤情况,酶联免疫吸附法检测肺泡灌洗液中炎症因子含量,蛋白质印记法检测损伤肺组织中丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)信号通路家族蛋白的表达情况。结果:不同剂量祖卡木能显著改善大鼠肺组织的炎性病理损伤,降低肺泡灌洗液中的炎症因子分泌水平,并抑制肺组织中MAPK信号通路家族蛋白的活化,其中高剂量祖卡木颗粒的改善效果和抑制作用最佳。结论:祖卡木具有显著的抗炎作用,其具体作用机制可能与抑制MAPK信号通路的活化和炎症因子的释放有关。Objective:To investigate the anti-inflammatory effects of traditional Uighur medicine Zukamu granule in rats with acute lung injury and its possible mechanisms.Method:Forty-eight rats were randomly divided into six groups:control group,model group,positive drug group,low,medium and high dose of zukamu group(n=8).Animals in drug treated group were treated with low,medium or high dose of zukamu(1.35,2.7 and 5.4 g/kg)by intragastric administration for 5 days,respectively,or treated with positive drug Dexamethasone(5 mg/kg)by intraperitoneal injection once at the last day.However,animals in the control group and model group were injected with the same volume of saline.After drugs administration,lipopolysaccharide was used to induce acute lung injury in rats.The pathological damages of lung tissues in rats were observed by hematoxylin-eosin staining.The levels of inflammatory cytokines in bronchoalveolar lavage fluid(BALF)in rats were measured by enzyme linked immunosorbent assay and the protein expression of mitogen-activated protein kinase(MAPK)signaling pathway related proteins in damaged tissues were detected by Western blot.Result:Different doses of Zukamu could significantly improve the inflammatory pathological damages of lung tissues in rats,decrease the levels of inflammatory cytokines in BALF,and inhibit the activation of MAPK signaling pathway related proteins in lung tissues.Among them,the high dose group had the best effect.Conclusion:Zukamu granule has obvious anti-inflammatory effects,which may be related to inhibiting the activation of MAPK signaling pathway and the release of inflammatory factors.
