人血浆中3-甲氧基肾上腺素和3-甲氧基去甲肾上腺素的萃取和浓度检测方法研究

摘  要:目的:建立固相萃取-高效液相-电化学检测器联用法测定人血浆中3-甲氧基肾上腺素(Metanephrine,MN)和3-甲氧基去甲肾上腺素(Normetanephrine,NMN)浓度的方法。方法:采用固相萃取弱阳离子交换小柱萃取人血浆中的MN和NMN,用HPLC-ECD测定其浓度,并对其方法专属性、回收率进行研究。结果:MN线性相关系数R^(2)=0.9972,NMN线性相关系数R^(2)=0.9996;MN的提取回收率和方法回收率为84.34%和99.10%,NMN的提取回收率和方法回收率为90.55%和100.48%。结论:本课题建立的固相萃取-高效液相色谱-电化学检测器联用检测法检测人血浆中MN和NMN的浓度,操作简单,提取率较高,为临床人血浆中MN和NMN浓度检测提供方法学参考。Objective:To establish a method of determining metanephrine(MN)and normetanephrine(NMN)concentrations in human plasma by solid phase extraction and high performance liquid chromatography with electrochemical detection.Methods:The Waters OASIS WCX Cartridge is used to separate MN and NMN in plasma,then MN and NMN are determined by HPLC-ECD.The specificity and recovery of the method are studied.Results:The correlation coefficient(R^(2))for MN and NMN are 0.9972 and 0.9996,respectively.The recoveries of extraction for MN and NMN are 84.34%and 90.55%,respectively.The recoveries of methods for MN and NMN are 99.10%and 100.48%,respectively.Conclusion:The established method by solid phase extraction and high performance liquid chromatography with electrochemical detection has high recovery and easily operation,and it is sutable for determining MN and NMN in human plasma.
