摘  要:目的探讨苯并[a]芘(BaP)对人羊膜上皮细胞水通道蛋白-8(AQP-8)表达的影响及其作用机制。方法分离培养人羊膜上皮细胞建立细胞模型进行机制研究。Western blot检测BaP处理的细胞中AQPs表达与相关信号通路分子表达。结果与对照组[二甲基亚砜(DMSO)溶剂]相比,人羊膜上皮细胞中AQP-8、蛋白激酶A(PKA)以及环磷酸腺苷(cAMP)随着BaP浓度的增加呈剂量依赖性降低(P均<0.01),细胞色素P4501A1(CYP1A1)随着BaP浓度的增加呈剂量依赖性升高(P均<0.01)。当加入cAMP信号通路激活剂毛喉素(Forskolin),AQP-8、PKA以及cAMP表达与BaP处理组相比均升高(P均<0.01)。结论环境污染物BaP通过cAMP-PKA信号通路影响人羊膜上皮细胞AQP-8表达,可能是羊水平衡失调的机制之一。Objective To investigate the effect and mechanism of BaP on the expression of AQP-8 in human amniotic epithelial cells.Methods Human amniotic epithelial cells were isolated and cultured to establish cell models for mechanism study.Western blot was used to detect the expression of AQPs and related signaling pathway molecules.Results Compared with the control group(DMSO),AQP-8,PKA and cAMP decreased in a dose-dependent manner with the increase of BaP concentration(P all<0.01),and cytochrom P4501A1(CYP1A1)increased in a dose-dependent manner with the increase of BaP concentration in hAECs(P all<0.01).When the cAMP signaling pathway activator Forskolin was added,the expression of the AQP-8,PKA and cAMP were significantly increased compared with that of the BaP treatment group(P all<0.01).Conclusion The environmental pollutant BaP affects the expression of AQP-8 in human amniotic epithelial cells through the cAMP-PKA signaling pathway,which may be one of the mechanisms of amniotic fluid imbalance.
